粗壮女贞苦丁茶对α-淀粉酶和α-葡萄糖苷酶活性的抑制作用

摘要
研究粗壮女贞苦丁茶对α-淀粉酶及α-葡萄糖苷酶活性的抑制作用。采用紫外可见分光光度法,评价了粗壮女贞苦丁茶对α-淀粉酶及α-葡萄糖苷酶活性的抑制能力,并以Caco-2细胞为模型,进一步观察了其在细胞层面的抑制作用;通过化学分析方法,测定了粗壮女贞苦丁茶的总酚和总黄酮含量、自由基清除活性、脂质过氧化抑制作用及还原能力。结果表明,粗壮女贞苦丁茶能有效地抑制α-淀粉酶及α-葡萄糖苷酶的活性,在Caco-2细胞模型中亦能显著地降低蔗糖酶和麦芽糖酶的生物催化活性。此外,粗壮女贞苦丁茶还具有较高的总酚和总黄酮含量及较强的还原能力,亦能有效地清除实验体系中的羟自由基和超氧阴离子自由基并抑制脂质过氧化反应的发生。研究工作为粗壮女贞苦丁茶资源在食品、医药、生物、日化等工业的高值转化利用提供了实验基础。
Theinhibitory activities of Ligustrum robustum(Rxob.) Blume extract(LRE) on o-amylase anda-glucosidase wereinvestigated.By using colorimetry testing,the inhibitory activities of LRE on a-amylase anda-glucosidase weredetermined.Furthermore,its inhibitory capacity was further observed in Caco-2 cells.Meanwhile,the total phenols and flavonoids contents,free radical scavenging ability,lipid peroxidation inhibition and reducing power were also evaluated by chemical analysis methods.All results showed that LRE possessed the remarkable ability to inhibit the activities of a-amylase anda-glucosidase,and exhibited the effective inhibition on the catalytic activity of sucrase and maltasein Caco-2 cell monolayer.Moreover,LRE had the excellent ability to scavenge free radicals(superoxide and hydroxyl radicals),inhibit the lipid peroxidation and also had strong reducing power,with the high contents of total phenols and flavonoids.Present study could provide the experiment basics for the valuable application of LRE in food,medicine,biologyand daily chemistry industries.
