耐热木聚糖酶产生菌的筛选及产酶条件优化

摘要
耐热木聚糖酶由于其温度稳定性好、应用范围广泛等特点,成为生物技术领域研究的新热点。本论文采用T_(top)三步筛选法,从山东淄博中高温酒曲中筛选得到一株产耐热木聚糖酶菌株Z3-2。对其发酵产酶条件进行单因素实验考察,结果显示:碳源粒度20-40目,碳源浓度4%(w/v),初始培养基pH=6.0,发酵温度50℃,转速200r/min时,该菌所产木聚糖酶活力高达5141.61 U/mL,较未优化前提高了2.7倍;对Z3-2菌株所产耐热木聚糖酶的酶学性质进行初步探究:该耐热木聚糖酶的最适pH为5.5且在pH=5.0-8.0时性质稳定,酶活保留达80%以上;Z3-2菌株所产耐热木聚糖酶最适于玉米芯木聚糖(纯度90%)的降解,其还原糖产量高达2.35 mg/mL,且产物以木二糖为主。综上所述,该耐热木聚糖酶在低聚木糖生产中具有一定的应用前景。
The heat-resistant xylanase is becoming a new research hotspot in the field of biotechnology because of its good temperature stability,and the features of wide range of applications.In this studuy.A xylanase producing strains named Z3-2were screened from the moderate-high distiller's yeast of ShanDong Zibo by T_(top) three-step approach.The single factor experiment results for fermentation condition showed as follows;mesh number of carbon source;20-40;carbon source concentration;4%;the initial medium pH;6.0,fermentation temperature;50℃,the rotate speed of shaker;200 r/min,under the optimum condition of the cultivation,the highest enzyme activitycan reach 5141.61 U/mL,2.73 times than before.Making a preliminary study on enzymatic properties secreted by Z3-2,the optimum pH = 5.5;Among the pH = 5.0-8.0,the xylanase is very stable,the enzyme activity reserve more than 80%;the xylanase is more fit for 90%corn cob xylan,hydrolysates is main xylobiose,production of reducing sugar reached 2.35mg/mL.In conclusion,the heat-resistant xylanase has a certain application prospect on xylooligosaccharides production.
