杆菌肽锌对断奶獭兔盲肠菌群和免疫功能的影响

摘要
为研究杆菌肽锌对断奶獭兔盲肠菌群和免疫功能的影响,本研究选用40只28日龄断奶獭兔,随机平均分为在基础日粮中添加杆菌肽锌(40 mg/kg)的实验组和不添任何抗生素的对照组。采用聚合酶链式反应-变性梯度凝胶电泳(PCR-DGGE)和荧光定量PCR(qPCR)方法对断奶獭兔盲肠菌群和TLR2、TLR4 mRNA表达水平进行检测,酶联免疫吸附试验(ELISA)对外周血细胞因子进行测定。PCR-DGGE结果表明,与对照组比较,实验组盲肠菌群丰富度、多样性和均匀度等没有显著变化(p>0.05)。qPCR检测表明,实验组的梭菌类群XIVa显著降低(p<0.05)。在免疫功能调节方面,实验组外周血细胞因子IL-1、IL-2、IL-4、TNF-α的浓度显著升高(p<0.05);TLR2、TLR4 mRNA的相对表达量总体呈上调趋势。以上结果表明,杆菌肽锌作为断奶獭兔饲料添加剂,对盲肠菌群结构没有显著影响,但能够增强机体免疫力。
This experiment was conducted to evaluate the effects of zinc bacitracin on weaned rex rabbits cecum microflora and immune function.A total of 40 rex rabbits weaned at 28 days of age were randomly allotted into two groups inclouded experimental group was fed the basal diet supplemented with zinc bacitracin 40 mg/kg and control group was fed with basal diet without any antibiotics.The cecum microflora,mRNA expressions of TLR2 and TLR4,and peripheral blood cytokines in weaned rex rabbit were detected by PCR-DGGE real-time PCR and ELISA,respectively.PCR-DGGE results showed that compared with control group,richness and evenness and microflora diversity were not significant changed in experimental group(p>0.05).Real-time PCR detection results showed that the Clostridium(cluster XTVa) in experimental group was greatly decreased(p<0.05).In terms of immune function,the cytokine concentrations in peripheral blood,such as IL-1,IL-2,IL-4 and TNF-α were increased significant in experimental group(p<0.05).The relative mRNA expression levels of TLR2 and TLR4 were up-regulated trends in the experimental group.These results indicated that use of zinc bacitracin as a feed additive to weaned rex rabbit might enhance immune abilities and have no significant effects on cecum microbial community structure.
