FT-IR技术快速鉴定硫酸盐还原菌

摘要
本文采用傅里叶变换红外光谱(FT-IR)法,研究了湛江(ZJ)和渤海(BH)油田污水中硫酸盐还原菌(SRB)的特征峰,并结合两株细菌的生理生化特性进行了菌种鉴定,研究结果表明结合标准菌株的FT-IR特征峰位置的吸光度,FT-IR法可快速分析并判断ZJ油田SRB属于脱硫脱硫弧菌属,BH油田SRB为脱硫肠状菌属。同时运用分子生物学方法聚合酶链反应(PCR)对SRB的16S rDNA基因特征性序列进行测序,鉴定结果与FT-IR的结果吻合。因此FT-IR可作为一种快速鉴定SRB类群的方法。
Fourier transform infrared spectroscopy(FT-IR) was applied to study the specific peaks of sulfate reducing bacteria(SRB) of Zhanjiang(ZJ) and Bohai(BH) oil field wastewater,combined with the physiological and biochemical features of these bacteria species,the SRB of ZJ oil was primarily determined as Desulfovibrio and the SRB of BH oil as Desulfotomaculum.On the other hand,molecular biological method of polymerase chain reaction(PCR) was utilized to characterize SRB by the feature sequences of 16 S rDNA gene,the results by PCR coincided with the results by FT-IR.In conclusion,FT-IR method can be used to rapidly and accurately characterize and identify SRB.
