富锌滑子蘑菌丝体和子实体中谷胱甘肽及其合成关键酶的研究

摘要
谷胱甘肽做为体内一种重要的抗氧化剂,能够清除掉人体内的自由基,提高人体免疫力。本论文研究了不同富锌浓度对滑子蘑的菌丝体和子实体中谷胱甘肽及其合成关键酶的影响,培养浓度为:0、200、400、600、800、1000mg/kg。采用从南京建成生物工程研究所购买的γ-谷氨酰半胱氨酸合成酶(γ-GCS)测定试剂盒测定酶活,四氧嘧啶法测定谷胱甘肽的含量。实验结果:菌丝体和子实体中的γ-GCS活性及谷胱甘肽含量随着富锌浓度的增加整体呈上升趋势且均大于相对应的空白组;子实体中的γ-GCS活性大于菌丝体中的γ-GCS活性,富锌浓度为1000 mg/kg时,菌丝体及子实体的γ-GCS具有最大活性,分别是1.87U/g、0.70 U/g;但菌丝体中的谷胱甘肽含量大体上高于子实体中的谷胱甘肽含量且都大于相对应的空白组,在富锌浓度为1000 mg/kg时,菌丝体及子实体的谷胱甘肽含量达到最大值,分别为347.0mg/g、320.3mg/g。进而得到结论,富锌对滑子蘑生长过程中谷胱甘肽的合成有明显的促进作用,可以进一步做大批量实验验证,继而为投入生产做准备。
Glutathione as an important antioxidant in vivo,can rid the body of free radicals,enhance human immunity.This paper studied the effect of different concentration of Zn-enriched Pholita Namek mycelium and fruiting body of its glutathione and key synthase,the training concentration as:0,200,400,600,800,1000 mg/kg.Using the 7-glutamyl cysteine synthetase(7-GCS) activity assay kit that purchased from Nanjing Jiancheng Bioengineering Institute to detect activity,the content of glutathione was measured by alloxan method.The results:γ-GCS activity and the content of glutathione in mycelium and fruiting body was in upward trend overall with the increasing concentration of zinc and each of them were greater than the corresponding blank group;γ-GCS activity in fruiting body was higher than that of mycelium,in the Zn-enriched concentration of 1000 mg/kg,the γ-GCS activity in mycelium and fruiting body having maximum value,namely 1.87 U/g,0.70 U/g;however the content of glutathione in mycelium is higher than it in fruiting body,and both of them were greater than the corresponding blank group,in Zn-enriched concentration of 1000 mg/kg,the content of glutathione in mycelium and fruiting body reached a maximum value,347.0μg/mg,320.3μg/mg.Further conclude that Zn-enriched on Pholita Namek growth process could have significant role in promoting about synthesis of glutathione,can further enlarge and experimental validation batches,and then prepare for the production.
