丁香有效部位抑制LDL中脂类及蛋白氧化修饰及光谱学变化

摘要
低密度脂蛋白(LDL)氧化修饰是致动脉粥样硬化(AS)的关键性因素。在氧化修饰过程中LDL中脂类、蛋白均会受到影响并改变LDL光谱学性质。实验室前期研究结果表明,丁香在国家公布的87种药食两用原料中抗氧化活性最强,并确定了其抑制LDL氧化修饰的有效部位。本试验运用紫外吸收光谱、荧光光谱等方法研究了丁香有效部位(Effective fraction from clove,EFC)对LDL氧化修饰的抑制效果。结果显示,EFC能有效延缓LDL氧化过程中共轭二烯(CD)增殖及推后其达到最大值;抑制胆固醇降解、色氨酸(Trp)荧光淬灭、赖氨酸(Lys)修饰和荧光吸收变化及脂褐素生成,且呈一定量效关系;影响LDL紫外光谱学特征的改变。结果表明EFC能有效抑制LDL氧化修饰。
Oxidation of low density lipoprotein(LDL) have been considered as the critical factor which led to atherosclerosis(AS).Lipid and protein that LDL contains were oxidized to cause change of spectra during oxidation.Clove has been demonstrated to possess the strongest antioxidant capacity among 87 kinds of both medicinal and edible raw materials released by the government,and the effective fraction on LDL oxidation inhibition was proved in the former study of our laboratory.The inhibition efficiency of the effective fraction from clove(EFC) on oxidation of LDL was studied by UV-Visible spectra and fluorescence spectra.The results indicated that EFC may effectively delay propagation of conjugated diene(CD) generated during oxidation of LDL and postpone to reach maximum of its content.EFC may inhibit cholesterol degradation,fluorescence of tryptophan(Trp) quenching,lysine(Lys) residues being modified,change of fluorescence spectra of Lys and lipofuscin generated during oxidation of LDL.These inhibition efficiency depended on concentrations of the effective fraction.EFC also may affect change of UV-Visible spectra of LDL during oxidation.The results suggested that EFC may effectively inhibit oxidation of LDL.
