策划药及其代谢物的分析方法研究

摘要
策划药是指对列管毒品进行化学结构修饰所得到的毒品类似物或衍生物,其具有与受管制毒品相似的兴奋或者致幻效果。该类药物种类较多,主要包括阿片类、致幻剂类、解离性麻醉剂类、哌嗪类、苯丙胺类兴奋剂、卡西酮类、合成大麻素、镇静剂类、类固醇类等9种,当今非法市场上较为流行的两种新型策划药为合成大麻素(香料)和卡西酮类(浴盐)。基于液相色谱-电喷雾离子阱质谱联用技术,我们已经建立了药物及其代谢物快速高通量分析方法。本文建立了亚甲二氧吡咯戊酮(MDPV)等策划药及其代谢产物的测定方法。借助LC-MS联用技术,在大鼠肝微粒体S9组分中鉴定得到四种策划药的5种代谢产物。研究结果表明:所建立的方法用于策划药分析,不仅可以节省分析时间、减少实验成本,而且具有高效、灵敏和选择性好等特点。
A sensitive liquid chromatography-electrospray ionization-ion trap mass spectrometry(LC-ESI-ITMS)method was utilized for the simultaneous analysis of four designer drugs and their in-vitro metabolites in rat liver microsome S_9 fraction. Four designer drugs, including methcathinone(MC), 3,4-methylenedioxymethcathinone(MDMC), 3,4-methylenedioxy-pyrovalerone(MDPV) and 4'-methyl-α-pyrrolidinopropiophenone(MPPP), were individually incubated with rat liver microsome S_9 fraction, and the incubation mixtures were pooled together and analyzed by LC-ESI-ITMS simultaneously. Five of their main metabolites were identified via the analysis of protonated molecules and tandem mass spectrometry data. The studies indicated that the approach could be applied in the determination of illicit drugs and their metabolites in medical, pharmaceutical and forensic investigations.
