摘要
建立HPLC法测定麻黄鱼腥草散中黄芩苷、穿心莲内酯、脱水穿心莲内酯含量的方法。色谱柱为Waters XBrige~(TM)C_(18),4.6×150mm,5μm;黄芩苷以甲醇-0.2%磷酸溶液为流动相梯度洗脱;流速:1.0mL/min;柱温:30℃:穿心莲内酯、脱水穿心莲以甲醇-水(50:50)为流动相;流速:1.0mL/min柱温:30℃;检测波长:黄芩苷检测波长276nm,穿心莲内酯检测波长225 nm,脱水穿心莲内酯检测波长为254nm。黄芩苷的线性范围为0.0798~0.7975μg,R=1.0000,平均回收率为94.3%(RSD=0.8%)。穿心莲内酯的线性范围为0.0109~0.2175μg,R=1.0000,平均回收率为92.9%(RSD=0.7%);脱水穿心莲内酯的线性范围为0.0105~0.2105μg,R=0.9999,平均回收率为92.4%(RSD=0.3%)。本方法快速、简便、准确,能有效测定麻黄鱼腥草散中黄芩苷、穿心莲内酯、脱水穿心莲的含量。
To establish an HPLC method for determination of baicalin,andrographolide and dehyandrographolideinMa Huang Yu Xing Cao powder.The baicalin was analyzed on an XBrige~(TM) C_(18) column(4.6 mm×250 mm,5 μm) using ultrapure methanol-0.2%phosphoric acid solution as the mobile phase.The flow rate was 1.0 mL/min.The column temperature was 30 ℃.The andrographolide and dehyandrographolide were analyzed on an XBrige~(TM) C_(18) column(4.6 mm×250mm,5 μm) using methanol-ultrapure water(50:50) as the mobile phase.The flow rate was 1.0 mL/min.The column temperature was 30℃.The detection wavelengthsofBaicalinwas set at 276 nm.The calibration curve was linear in the ranges of 0.0798~0.7975 μg(R = 1.0000) for baicalin.The recovery of baicalin was94.3%with RSD of 0.8%.Andrographolide and dehyandrographolide were detected at 225 nm and 254 nm.The calibration curves were linear in the ranges of 0.0109~0.2175 μg(R = 1.0000) and0.0105~0.2105μg(0.9999) for andrographolide and dehyandrographolide.The average recoveries were 92.9%for andrographolide(RSD=0.7%,n=6) and 92.4%for dehyandrographolide(RSD=0.3%,n=6),respectively.This method is rapid,simple and accurate,which can be used for the determination of baicalin,andrographolide and dehyandrographolidein Ma Huang Yu Xing Cao powder.
