α-L-鼠李糖苷酶转化芦丁及酶固定化的研究

摘要
异槲皮苷是芦丁的衍生物,结构上比芦丁少一个鼠李糖,是一种具有比芦丁更高的药理活性的黄酮类化合物,因而将芦丁转化为异槲皮苷在食品及医药产业上有重大意义。本研究通过α-L-鼠李糖苷酶转化芦丁制备异槲皮苷,其中最适反应温度为60℃、最适反应pH为4.0。在最优反应条件下进行酶反应并利用HSCCC对产物进行分离纯化,得到180 min至220 min之间所收集的产物是异槲皮苷;本研究还利用11%PVA与0.5%海藻酸钠作为载体,加入浓度为2%的戊二醛交联1h后,加入酶液445.73 U,振荡吸附2h,挤入4%硼酸与1%CaCl_2混合溶液中固化15 min,酶活回收率为32.88%。固定化的α-L-鼠李糖苷酶最适反应温度为35-50℃,最适反应pH为4.0,该固定化酶在连续使用6次后酶活仍然保留98.22%,具有良好的重复利用性。实现酶法转化芦丁制备异槲皮苷及固定化α-L-鼠李糖苷酶对α-L-鼠李糖苷酶在食品和医药领域的应用有重大意义。
Isoquercitrin is a kind of flavonoids pharmacological,a derivative of rutin,which is less one rhamnose than rutin in structure but has more effective than rutin in activity.Therefore,it have great influence on bioconversion of rutin to isoquercitrin in food and medicine field.Thisstudy aimed to convert rutin into isoquercitrin using recombinant ot-L-rhamnosidase.The optimum temperature and pH of reaction wereat 60℃ andpH 4.0.HSCCC was used to purify the product,and the productof isoquercitrinwas collected between 180 min to 220 min when the peak came out.Finally,made use of 11%polyvinyl alcohol(PVA) and 0.5%sodium alginate to immobilize ot-L-rhamnosidase into matrix and the matrix would be crosslink with 2%glutaraldehyd for 1h and loaded with enzyme with total activity of 445.73 U,2h of absorption time,4%boric acid,1%CaCl_2,respectively.The recovery rate of the enzyme was 32.88%.The optimum temperature of reaction was 35-50℃,and the optimal pH of reaction was 4.0.The immobilized ot-L-rhamnosidase still retained over 98.22%of its original activity after 6 cycles reusing,indicated its good quality of reusing.Therefore,it is of great significance in the application of ot-L-rhamnosidase in food and medicine field.
