明胶/氧化石墨烯纳米复合材料固载葡萄糖氧化酶

以天然鳞片石墨为原料,采用改良Hummer法制备了单分散氧化石墨烯(GO),对其化学组成、晶态结构和形貌进行了系统表征和分析,证实产物具有单层结构,表面含有大量含氧基团,在水中具有良好的单分散性;以医用明胶和自制GO为原料,采用水相简单自聚合方式制备了明胶/GO纳米复合膜,对其进行了系统表征和性质测定,表明GO在明胶载体中分散均匀、良好,添加0.5wt%GO的纳米复合膜性能较好,GO能有效改善明胶的机械性能及热稳定性;以明胶/GO纳米复合膜为载体,戊二醛为交联剂,固定葡萄糖氧化酶,对固定化条件及固载酶的多项性质进行了研究,确定酶的理想固定条件为1 cm~2明胶/GO纳米复合膜(厚度约为100.μm)与5mL 4%戊二醛交联,固载酶液最佳pH 6.0,2 h可完成固载,可固载6mg葡萄糖氧化酶;固定化酶的最适温度为50℃,最适pH6.0,动力学参数Km值为28.24mmol/L,较游离酶Km值略高,其热稳定性明显提高,具有良好的操作及贮存稳定性。与单纯明胶载体相比,明胶/GO纳米复合膜可在较短时间内实现更大固载量,能提高葡萄糖氧化酶的热稳定性,酶活无明显降低,是一种性能良好的新型固载材料。
Using natural flake graphite as raw material,the single dispersed graphene oxide(GO) is prepared by modified Hummer method.Characterizations and analyses of the chemical composition,crystal structure and microstructure show the product is with a single layer structure,contains a large number of oxygen containing groups on its surface,has good monodispersity in water.Using medical gelatin and self-made GO as raw materials,gelatin/GO nanocomposite membrane is prepared by simple self-assembly of both components in aqueous media.It is observed that GO is dispersed on a molecular scale in the gelatin matrix and some interactions occur between gelatin and GO sheets.Compared with pure gelatin,mechanical behavior and heat stability of nanocomposite membrane can be significantly improved by 0.5wt%GO loading.Glucose oxidase is immobilized on gelatin/GO nanocomposite membrane with glutaraldehyde by cross-linking reaction,the immobilization conditions and characterization of the immobilized enzyme are studied.The ideal conditions for immobilization are as follows:1 cm gelatin/GO nanocomposite membrane(is treated with 5mL 4%glutaraldehyde,best acidity of enzymic solution is pH6.0,than 6mg glucose oxidase can be immobilized on to the carrier in 2h.Optimal temperature and pH for the immobilized enzyme are 50℃ and6.0 respectively.Kinetic parameter,Km is 28.24mmol/L by Iineweaver-Burk method.The temperature stability of the immobilized enzyme is better than free emzyme,and the immobilized enzyme also shows good operation and storage stabihty.Compared with the pure gelatin carrier,gelatin /GO nanocomposite membrane is a new type of good performance of matrix materials for it can load more enzyme in shorter time,improve the thermal stability of glucose oxidase but not reduce the enzyme activity.
