中国野生蓝莓花青素对不同食源性致病菌的抑菌机理研究

蓝莓中富含花青素等多酚类物质。花青素的生物活性显示了其在预防慢性退化疾病方面的潜在可能。为了了解花青素如何抑制食源性致病菌的生长,本文研究了中国野生蓝莓对单增李斯特菌,金黄色葡萄球菌、肠炎沙门氏菌、副溶血性弧菌四种食源性致病菌的抑菌作用。结果显示花青素对单增李斯特菌,金黄色葡萄球菌、肠炎沙门氏菌、副溶血性弧菌的最小抑菌浓度分别为0.53、0.27、0.53、0.13 mg/mL。扫描电镜发现花青素能够破坏菌体的原有形态,致使胞内物质泄漏。当花青素作用细菌两小时后,细菌总蛋白含量减少,AKP酶、ATP酶活性分别减少54.3%,69.7%。随着花青素浓度的不断增加,OD630明显降低,细菌代谢能力减弱,继而影响了细菌的能量转换,抑制了其生长及再生。花青素通过破坏细胞膜及其生长环境,影响细菌的新陈代谢,降低了细菌活性。本研究表明中国野生蓝莓花青素具有较强的抑菌活性,可以在预防医学、食品安全等领域加以应用,并丰富了人们对于野生蓝莓有利于健康的认识。
Blueberries are rich in polyphenols such as anthocyanins.Anthocyanins are nutraceuticals which show the potential to prevent chronic degenerative diseases through their biological activities.The objective of this study was to evaluate the anthocyanins how to inhibit the growth of foodborne pathogens.The antibacterial effects of Chinese wild blueberry anthocyanins on 4pathogenic bacteria,L.monocytogenes,S.aureus,S.enteritidls and V.parahaemolyticus,had investigated.The minimum bactericidal concentrations(MBCs) of anthocyanins against L.monocytogenes,S.aureus,S.enteritidis,V.parahaemolyticusrespectively were 0.53,0.27,0.53,0.13 mg/mL.The scanning electron microscopy micrographs shows that anthocyaninsdestroyed the original shape of the cell,so that intracellular material leak.When the bacterias treated with anthocyanins for2 hours,total protein content decreased in bacteriaand the enzyme of AKP,ATP decreased 54.3%,69.7%respectively。With increasing concentration of anthocyanins,the OD_(630) distinctly decrease which shows that bacterial metabolism was decreased,affecting the energy transfer of bacteria and inhibiting their growth and reproduction.Anthocyaninsaffected bacterial metabolismand reduced the bacterial cell activitythrough destroy the cell membrane and the growth environment of bacteria.The Chinese wild blueberry anthocyanins which had strong antibacterial as shown in this study could be applied to the field of preventive medicine,food safety,and enrich the understanding of the health benefits of wild blueberry.
