高效液相法同时测定抗肿瘤三棱复方提取液中4种化学成分

摘要
目的建立高效液相方法同时测定抗癌中药复方(三棱、制大黄、川芎)中4种化学成分,对其水提物和醇提物中各主要成分的含量进行对比,为优化复方提取工艺提供初步依据。方法色谱柱为Zorbax SB-C18(4.6×12.5mm,5μm),流动相为甲醇-0.1%磷酸水,梯度洗脱,分析时间18 min。利用所建立的方法对不同提取批次的水提物(3批)和醇提物(3批)进行含量测定。结果 4种化学成分的峰面积和浓度在测定浓度范围内均具有良好的线性关系,精密度RSD为3.6~8.5%,平均加样回收率为91.2~103.6%。结论本测定方法简便、灵敏度高、专属性好,适用于复方提取液的含量测定。
Objective: To create HPLC approach and determination of Common Burreed Rhizome compound(Common Burreed Rhizome, Prepared Rhubard, Chuanxiong Rhizoma) extractof 4 chemical composition,the water extract and alcohol extract content of each ingredient in the comparison, provide a preliminary basis for optimization of complex extraction process. Method: Chromatographic column for the Zorbax SB- C18(4.6 x 12.5 mm, 5 microns), mobile phase of methanol to 0.1% phosphoric acid water, gradient elution, 18 min analysis time. Using the approach ofdifferent batch of aqueous extract(3 batchs) and alcohol extract content determination(3 batchs). Result: 4 kinds of chemical composition of the peak area and concentration in the determination ofconcentration range has a good linear relationship, precision RSD was 3.6 ~ 8.5%, the recovery is 91.2 ~ 103.6% on average. Conclusion:This determination approach is simple, high sensitivity, good specificity, and is suitable for content determination of the compound extract.
