甘草次酸脂质体的制备与性质研究

摘要
目的:优化甘草次酸(GA)脂质体的工艺配方并考察其体外应用性能。方法:采用溶剂-超声法制备GA脂质体,以粒径、稳定性为综合评价指标,优化制备工艺及配方,扫描电镜观察脂质体的粒径形态及大小。利用高效液相色谱法测定GA含量,进行体外释放和透皮实验的表征。结果:GA脂质体最佳工艺处方为超声时间2min;超声功率30%;吐温-80加入量0.33%,氯化钠加入量为0.05%。制备的脂质体粒径均匀,呈球形小囊泡。分散性好,粒径(117±4.64)nm,包封率(91.0±1.21)%;GA脂质体具有明显的缓释效果,可有效作用于真皮层中;GA脂质体在4℃的存放条件下,30d内粒径变化较小,稳定性良好。
Purpose:Optimization the formula of Glycyrrhetinic acid(GA) liposome and investigate its application performance in vitro.Mehtods:Solvent-ultrasonic method was used to prepare GA liposome;Using particle size and stability as the comprehensive evaluation index to optimize the preparation technology and formula;Scanning Electron Microscope was used to observe the particle morphology and size of liposomes.The GA content was determined by High Performance Liquid Chromatography(HPLC) method for the characterization of the in vitro release and transdermal experiments.Results:The optimum process recipe for GA liposome was as follows:the ultrasonic time was 2 min;the ultrasonic power was 30%;the Tween-80 content was 0.33%and the amount of sodium chloride was 0.05%.The prepared liposome was of uniform particle size and present spherical capsule.The particle size was(117±4.64) nm,the encapsulation efficiency was(91.0± 1.21)%.GA liposome had obvious sustained release effect and can be effectively act in the dermis.Under the storage condition of 4 ℃,the particle size GA liposome was rarely changed within 30 days which demonstrated he stability was good.
