木聚糖酶筛选及其催化生成益生元型特异水解产物性质研究

以木聚糖制取有较高附加值的功能低聚糖是富含半纤维素材料的农业废弃物再生利用的有效增值途径,具有巨大的潜在经济效益和环保意义。研究对前期筛选的一株产木聚糖酶L10608菌株进行形态学及分子生物学鉴定,判定其为链霉菌。对该菌株所产木聚糖酶通过30%-50%硫酸铵沉淀,SP-sepharose离子交换树脂交换层析,纯化得到电泳级纯度木聚糖酶L10608-Xyn11。通过SDS-PAGE测定其酶蛋白质分子量为24 kDa。探究L10608菌株所产木聚糖酶以商品玉米芯木聚糖、商品燕麦木聚糖、实验室自提水不溶性玉米芯木聚糖及为底物时的水解特性,通过高效液相色谱检测其水解底物的组成及比例,结果表明该菌所产木聚糖酶在水解玉米芯木聚糖时,对木三糖有很强的水解作用,水解产物主要为木二糖,伴有少量木糖产生。该酶对木二糖的基本无水解能力。菌株L10608产酶水解特异性表明该菌有望作为益生元型低聚木糖的生产菌株。
Preparation of oligosaccharide from xylan hydrolysis is an effective way to add value to hemicellulose material of agricultural waste recycling,which has enormous economic potential and environmental protection.Study of a xylanase producing pre-screening strain L10608,determiningby morphological and molecular biology identification as Streptomyces.A 24 kDa xylanase from StreptomycesL10608 was purified and its hydrolysis products analyzed by HPLC.The xylanase was purified to homogeneity by 30%-50%ammonium precipitation and anion-exchange chromatography.The xylanase was hydrolyzed and analyzed by three different substrates.The xylanase displayed strong hydrolysis ability on xylotriose but weak on xylobiose,results in hydrolyzate mainly is xylobiose,indicating that the enzyme had potential application prospect in prebiotics production.
