响应面法优化花生壳中木犀草素提取工艺研究

摘要
为利用花生壳中有效成分木犀草素,研究木犀草素最佳提取工艺条件并测定其含量。在提取溶剂浓度、料液比、时间、温度单因素实验基础上,根据中心组合设计原理采用三因素三水平的响应面分析法进行最佳提取工艺的优化,并采用HPLC法测定木犀草素含量。分析各个因素的显著性和交互作用后,获得优化提取提取工艺条件为:81.6%甲醇,1:26.75料液比,2.3h提取时间。在该提取条件下,木犀草素得率为30.64μg/g。通过优化提取工艺条件,为充分利用花生壳有效成分提供基础。
The aim of this work was to study optimization extraction parameters of luteolin from peanut shell,and determine its content.The optimized extraction parameters was yield with central composite design by three factors three levels of response surface analysis,on the basis of single factor experiments including solvent concentration,ratio of solid to liquid as well as extraction time.The effects of process parameters were studied by Box-Behnken center united experiment designed by response surface methodology.The content of luteolin was determined by HPLC.Optimum yield of luteolin was 30.64μg/g,with81.6%methanol at 1:26.75 solid to liquid ratio for 2.3h.This work provides the reference and scientific basis for the use of peanut cultivate resource.
