摘要
口腔鳞癌是头颈部较为常见的肿瘤之一,~(1,2)近年来口腔鳞癌发病呈年轻化态势增长。由于传统的治疗药物存在明显的毒副效应,从而限制其在临床中的根本应用。因此,找寻一些新的治疗策略去弥补现在治疗方法的不足,是当前口腔医学研究领域的热点。本文利用静电纺丝技术,成功制备出载药纳米纤维膜,并对口腔癌细胞进行体外作用,系统研究了该载药缓释体系的抗癌效果。研究表明,载入顺铂药物并引入氯喹作为自噬抑制剂的聚乳酸静电纺丝膜,与正常使用药物作用CAL-27细胞相比,缓释体系具有更强、更持久的杀伤作用,同时具备良好的生物相容性。
Poly(L-lactide)(PLA) nanofibers containing the antitumor drug cisplatin/chloroquine were prepared using a one-step method by means of electrospinning of PLA solutions. The pristine and cisplatin/chloroquine-containing nanofiber mats were characterized by scanning electron microscope(SEM). SEM observation revealed that the diameter of the cisplatin/chloroquine-containing nanofiber fibers ranged from 450 nm to 650 nm. Furthermore, compared with raw cisplatin, the prepared drug-containing nanofibers showed an initial low antitumor activity and then provided a sustained antitumor efficiency. Therefore, the performance of these nanofibers makes them promising candidates for tumor treatment.
