超声-酶辅助提取银杏叶多糖

摘要
本实验以秋后银杏落叶为原料,以提取率及粗糖中的多糖含量为指标,通过超声处理及添加纤维素酶为辅助手段,经单因素实验及响应面实验提取银杏叶多糖,提取工艺如下:在功率为810w的条件下超声处理20min,再添加0.75%的纤维素酶,以蒸馏水为溶剂冷凝提取后加乙醇过夜,将沉淀冷冻干燥后得银杏叶粗多糖,测得其提取率为12.2%,经苯酚+硫酸法测得的粗糖中银杏叶多糖含量为72.38%。与传统的水溶醇沉法相比,提取率提高了1.1%,多糖含量提高了35.08%,提取时间减少了1h。本研究工艺简单效率高,为银杏叶多糖多糖的工业化制备提供了新的思路和方法。
In this experiment,Ginkgo leaves picked in autumn as raw material,raw sugar and the extraction rate of polysaccharide content as indicators,preparation of polysaccharide of single factor experiment and response surface experiment were carried out by water extraction and ethanol precipitation with sonication and cellulase as a secondary way.Polysaccharides extraction process of Ginkgo biloba leaves is as follows:the sonication power of 810 w for 20 min,then adding 0.75%cellulase,ethanol distilled water as the solvent condensate extracted overnight,the extraction precipitate was freeze-dried to obtain crude Ginkgo biloba polysaccharide.12.2%of extraction rate was measured and crude 72.38%of sugar polysaccharides was obtained by phenol-sulfuric acid measured.Compared with the conventional water-soluble alcohol precipitation method,the extraction rate increased by 1.1%,the polysaccharide content increased by 35.08%,extraction time decreased lh.In this study,So simple and high efficiency process provides a new idea and method for industrial preparation of ginkgo biloba polysaccharides.
