非洲菊组织培养和植株再生

摘要
非洲菊(Gerbera jamesonii)属多年生草本观赏植物,是世界十大鲜切花之一。本实验以课题组选育的非洲菊新品种‘幻彩'和‘火焰',商业品种‘高山'和‘革命红'以及课题组培育的杂交新品系HN-01为试材,进行非洲菊组织培养研究,探究适于不同非洲菊品种的最适培养基和培养条件。结果表明:最适宜非洲菊诱导愈伤并出芽的外植体是花托,‘火焰'芽诱导最适培养基是MS+BA 7.0 mg/L+KT 3.0 mg/L+NAA 0.3mg/L;‘幻彩'芽诱导最适培养基是MS+BA 7.0 mg/L+NAA 0.3 mg/L。光质对‘高山'和‘革命红'非洲菊的增殖培养具有显著影响。0.1 mg/L的NAA更适于非洲菊杂交新品系HN-01生根培养。
Gerbera jamesonii is a perennial herbaceous ornamental plant.It is one of the most important ten fresh cut flowers in the world.The tissue culture of new varieties of Gerbera which breeding in our research group G.jamesonii' Huancai',G.jamesonii ' Huoyan' and commercial varieties G.Jamesonii ' Gaoshan',G.jamesonii ' Revolution Red' and hybrid lines HN-01 were studied.The optimum culture medium and culture condition were explored in different varieties of G.jamesonii.It showed that the most suitable explanl for callus induction and Shoots Induction of Gerbera was the receptacle.The most suitable medium for the Shoots induction of ' Huoyan' was basal MS medium supplemented with BA 7.0 mg/L,KT 3.0 mg/L and NAA0.3 mg/L The most optimum medium for the Shoots induction of ' Huancai' was MS medium supplemented with BA 7.0 mg/L and NAA 0.3 mg/L Light quality plays Significant effect on the growth of G.Jamesonii ' Gaoshan' and G.jamesonii ' Revolution Red' during the proliferation.It showed that 0.1 mg/L NAA was more suitable for Gerbera hybrid lines HN-01 root culture.
