海藻多酚制备工艺优化及抗氧化活性

以大连海带,大连冻干海带,福建海带丝,福建海带结,大连裙带菜孢子叶为原料,以加热温度(25,50,75,100,115,125,150,200℃),加热时间(1,2,4,6,8h),提取溶剂(乙醇浓度分别为25,50,75,100%)为单因素,确定其最优条件;应用福林酚法测定海藻多酚的含量;应用响应面实验进一步优化裙带菜孢子叶海藻多酚的提取工艺条件,最后应用ABTS法测定5种原料海藻多酚的抗氧化活性。结果表明裙带菜孢子叶海藻多酚的最优提取条件为:加热温度170℃,加热时间5.2h,乙醇浓度52%,得裙带菜孢子叶海藻多酚的提取率为10.7mg/g(多酚/原料重)。以单因素的最优条件提取其他4种原料的海藻多酚,测定其抗氧化活性,结果表明,这四种海藻多酚提取率分别为:15.10,15.12,14.29,14.69mg/g;抗氧化活性分别为0.29,0.35,0.30,0.30mmol/gTE。优化的5种原料海藻多酚提取工艺对海藻多酚的生产制备提供了一定的理论参考依据,对开发功能性食品剂料奠定了一定的理论基础。
Dalian kelp,Dalian lyophilized kelp,Fujian kelp slices,Fujian kelp kink and Dalian sporophyll of Undaria pinnatifida(U.pinnatifida) were as raw material.The heating temperature(25,50,75,100,115,125,150,2001),heating time(1,2,4,6,8h),and extraction solvent(ethanol concentrations of 25,50,75,100%) were as a single factor to determine the optimal conditions;the phlorotannin content was measured by Folin- Ciocalteu method;application of response surface experiments to further optimize prepared conditions for phlorotannin,and the five kinds of raw seaweed phlorotannin antioxidant activity were measured by ABTS method.The results indicated that the optimal conditions of sporophyll of U.pinnatifida were as follows;heating temperature was 170℃,heating time was 5.2h,ethanol concentration was 52%,and sporophyll of U.pinnatifida phlorotannin extraction rate of 10.7mg/g(phlorotannin/raw material weight).The other 4 kinds of phlorotannin were prepared in optimal conditions and their antioxidant activities were measured,the results indicated that the 4 kinds of seaweed phlorotannin extraction rates were;15.10,15.12,14.29,14.69mg/g;antioxidant activities were 0.29,0.35,0.30,0.30mmoL/g TE.The optimal conditions of polyphenol extraction process for 5 kinds of seaweed provide some theoretical for the preparation process of phlorotannin,and provide the development of functional food material for a theoretical foundation.
