壳寡糖体外模拟胃消化机制研究

摘要
壳寡糖是自然界唯一一种天然阳离子寡糖,具有抗氧化、抑菌、抗肿瘤、调节血糖、降低胆固醇等多种重要生理活性,被列入新食品原料。然而壳寡糖在人体消化系统中的消化机制尚不明确,影响了对壳寡糖生理活性作用机制和营养功能的认知。本研究采用体外胃消化模型,探究了壳寡糖在模拟胃消化环境下消化过程的分子变化及与其他食物组分的分子相互作用,进一步探讨了上述变化和作用机制。结果表明,随着消化反应的不断进行,胃液中的酸和酶都会使壳寡糖发生不同程度的降解,壳寡糖的平均分子量以及总糖含量都下降,壳二糖,壳三糖的含量升高,壳五糖及以上聚合度的壳寡糖含量降低。此外,在消化过程中有类黑素物质和其它有机产物生成,这可能是发生了美拉德反应。本研究为壳寡糖在人体中的实际应用提供了有力帮助。
Chitooligosaccharides,listed as a new food material,is the one and only naturally cationic oligosaccharides,which have a variety of biological activities including antioxidant,antibacterial,anticancer,regulating blood glucose,cholesterol and so on.However,the digestive mechanism of chitooligosaccharides in human digestive system is not clear and this hinder the cognition of the nutritional function and the mechanism of physiological activities of the chitooligosaccharides.In this study,we use in vitro gastric digestion model to explore the molecular changes and interactions with other components of food during the digestive process,and further discuss the changes and mechanism of action during simulated gastric environment.The results show that along with the continuous digestion reaction,the acid and enzyme in gastric juice both play a critical role in the degradation of chitooligosaccharides at different levels.The average molecular weight and the tutal sugar content both decrease,the content of chitobiose and chitotriose increased while the content of DP 5-10 chitooligosaccharides reduced.In addition,melanoidin and some other organic products generated which might associated with Maillard reaction during the digestive process.This study provided help in actual application of chitooligosaccharides in human body.
