TR信号介导的四溴双酚A的脑发育毒性研究

摘要
TBBPA是全球应用最广的溴化阻燃剂之一,主要应用于环氧树脂的加工、电子设备和建筑材料的生产,并在环境介质和人体样本中均有检出[1]。它的化学结构与甲状腺激素(TH)相似,可以通过干扰TH与其受体(TR)的结合而干扰下游TH响应基因(TH-response gene)的表达。脊椎动物脑的发育受TR信号介导的TH的调控。如果污染物具有TR信号干扰作用,可能会通过这一途径影响脑的发育。两栖动物非洲爪蟾是研究TR信号的模式生物,故而使用非洲爪蟾研究TR信号介导的TBBPA的脑发育毒性。实验使用52期非洲爪蟾蝌蚪研究TBBPA对T3诱导的脑发育的影响,58期蝌蚪研究TBBPA对变态高峰期脑发育的影响。初步结果发现,TBBPA显著抑制T3诱导的脑中ST3、TH-bzip、MMP13等TH响应基因的表达。相应地,TBBPA抑制T3诱导的脑的形态学的重塑。
TBBPA is one of the most used brominated flame retardants,in epoxy resin processing or electronic equipment and building materials production.TBBPA was detected in environmental media and human samples[1].TBBPA has similar chemical structure with TH,and it can bind TR to disrupt expression of TH-response genes.TH regulate brain development of vertebrates,so TBBPA may have toxic effect on vertebrate brain development.In this study,we investigated the effects of TBBPA on brain development during T3-induced and spontaneous Xenopus laevis metamorphosis.We found that TBBPA significantly inhibited T3-induced transcription of TH-response genes,like ST3,TH-bzip,and MMP13.Correspondingly,TBBPA inhibited T3-induced brain remodeling at the morphological level.
