氧化纳米碳球在SALDI-TOF MS中的应用

摘要
基质辅助激光解吸飞行时间质谱(MALDI-TOF MS)因其高通量、高灵敏度等优点而成为蛋白、多肽及代谢组学研究中的有力工具~([1-2])。然而,常用的CHCA、DHB等有机基质在分子量低于500 Da时存在严重的背景干扰,限制了MALDIMS在小分子化合物检测中的应用~([3])。近年来,研究者~([4])提出使用无机纳米粒子或者纳米结构替代有机基质(SALDI-TOFMS),可以有效避免基质背景干扰,用于低分子量化合物分析。本文制备了形貌规整的纳米氧化碳球,使其作为MALDI基质成功运用于多种结构的小分子化合物分析。相比于石墨烯、碳纳米管等碳基材料,氧化纳米碳球制备方法简单、成本低廉。并且其良好的水分散性以及均匀的靶板分布形态,使得质谱检测的背景干扰小,重复性好,灵敏度高。
Oxidized carbon nanospheres(OCS) were prepared and demonstrated to own continuous π-conjugated structures,uniform nanosize and excellent water-dispersibility.Owing to their unique properties,the OCS was for the first time applied as an assistant matrix in Surface-assisted laser desorption/ionization time-of-flight mass spectroscopy(SALDI-TOF MS) for small molecules analysis.Compared with conventional organic matrix and other carbon-based matrices,low background interference and enhanced signal intensities were obtained by OCS matrix The good reproducibility and excellent salt tolerance of OCS matrix were also investigated.
