古尼虫草富硒发酵条件优化

本研究利用液体发酵技术,对古尼虫草进行富硒发酵,开发新的有机硒化物食品添加剂。以菌丝生物量及富硒率为参考指标,首先,对古尼虫草富硒发酵中最佳硒浓度进行筛选,结果得出,富硒发酵中亚硒酸钠的最佳浓度为7μg/mL;其次,利用单因素试验和正交试验对古尼虫草富硒发酵营养条件和培养条件进行优化,结果得出:最优发酵条件为蔗糖6%,蛋白胨0.6%,KH_2PO_40.3%,MgSO_4 0.25%,培养基pH 6.0,培养温度25℃,接种量8%;最后,对优化所得的发酵条件进行了验证试验,得出古尼虫草生物量为11.657±0.134g/L,富硒率为78.33%士0.67%,与对照组相比(11.067±0.224g/L,69.30%±0.43%)分别提高了5.32%和13.03%。亚硒酸钠在一定浓度下有毒性,培养基中亚硒酸钠浓度的不同将直接影响菌株的生长和富硒能力;液体发酵的培养条件和营养条件,对发酵过程中菌体的生长代谢有直接影响。有研究显示,蛹虫草、酵母、黑木耳富硒发酵中硒的浓度分别为10μg/mL,35μg/mL,50μg/mL。本研究为开发新的有机硒化物食品添加剂提供了又一有效途径。
To develop new food additives of organic selenium,Cordyceps gannii was fermented with selenium by using liquid fermentation.The biomass and rate of selenium-enriched were used as indicators.The optimal concentration of selenium in fermentation was screened.The results indicated that the optimal concentration of sodium selenite was 7μg/mL.Then the single factor test and orthogonal test were applied to optimize the conditions of fermentation.The optimal fermentation conditions were as follows:sucrose of 6%,peptone of 0.6%,KH_2PO_4of 0.3%,MgSO_4 of 0.25%,the initial pH of pH 6.0,culture temperature of 251,inoculum size of 8%.After the fermentation conditions were optimized,the biomass and rate of selenium-enriched were 11.657 ± 0.134g/L and 78.33%± 0.67%,respectively,which were increased by 5.32%and 13.03%compared to the control group(11.067 ±0.224g/L and 69.30%±0.43%).Due to the sodium selenitehas toxicityin toxicity,the concentration ofselenium in culture medium have direct influence on the growth and capacity of selenium enrichment of strain.The culture conditions and nutritional conditions of liquid fermentation also have direct influence on thegrowth and metabolism of the fermentation strain.The literatures indicated that the sodium selenite concentration in selenium-enriched fermentation of Cordyceps military,yeast,agaric were 10μg/mL,35μg/mL,50μg/mL,respectively.This study provided another effective way to develop new food additive of organic selenium.
