野茉莉(Styrax japonicus)组培快繁技术研究

摘要
本试验以野茉莉种胚苗为材料,对其进行离体快速繁殖研究。结果表明:用2g·L~(-1)NaClO处理低温贮藏的野茉莉种子15min即可达到较好的杀菌效果。增殖效果良好的培养基是MS+BA 0.1mg·L~(-1)+IBA0.3mg·L~(-1),增殖系数达到5.55,芽多且粗壮;采用两阶段诱导野茉莉生根,100mg·L~(-1)IBA处理7d,转入1/2MS培养,生根数最多,生根率为77.8%。泥炭土和珍珠岩比例为1:1作为基质,以穴作为容器,组培苗移栽成活率达到80%,组培苗叶色鲜艳,长势良好。该研究为品种的大规模繁殖和大面积推广提供了参考。
Experiments on tissue culture of Styrax japonicus was conducted by taking the seeds as explants.The results show that the 2g · L~(-1) NaClO handing of seeds that being cryogenic storage was better,the best soak time was 5 min.MS medium with0.1 mg · L~(-1) BA and 0.3mg · L~(-1) IBA was the most suitable for the propagation of Styrax japonicas,and the propagation rate reached 5.55 times,bud is abundant and healthy.Treatments with 100 mg/L IBA for 3 days had the best root induction effect,topped to 77.8%of rooting rate.Taking nutrition pot as container,peat soil and perlite as transplanting medium(1:1),the transplant survival rate was over 80%.There were many lateral roots and the plantlets which grew rapidly and vigorously.This method was suitable for tissue culture of Styrax japonicus and laid foundations for its large-scaled propagation and large area promotion.
