有机溶剂法提取鱿鱼内脏油脂及脂肪酸分析

鱿鱼内脏含有丰富的粗脂肪和蛋白,其粗脂肪含量占内脏干重20%~30%左右,其中不饱和脂肪酸约占65%,EPA和DHA占总脂肪酸32%左右,鱿鱼内脏是提取EPA和DHA的良好原料。本文采用不同方法提取秘鲁鱿鱼内脏的油脂,用气相色谱-质谱联用技术对提取油脂进行脂肪酸组成分析。采用有机溶剂、酶解、碱水解这三种方法对鱿鱼内脏的油脂进行了提取,提取条件优化及综合分析,最终采用有机溶剂提取法。有机溶剂法采用了氯仿-甲醇法、乙醇法、正己烷法,三种方法的平均提取率分别为60.61%、17.71%、22.72%。对不同方法提取的油脂进行脂肪酸分析,结果表明其中单不饱和脂肪酸含量为26.24%,多不饱和脂肪酸含量为39.01%。多不饱和脂肪酸中,EPA含量为7.14%,DHA含量为24.97%。鱿鱼内脏油脂中的不饱和脂肪酸含量丰富,成分与鱼油很相似。不同溶剂法提取对于鱿鱼油脂的脂肪酸含量影响不显著,但对其提取出的脂肪酸异构体种类影响较大。正己烷法能提取出的脂肪酸异构体种类最多,其次是酶+正己烷法,再次是氯仿-甲醇法,乙醇法最少。本文研究结果为鱿鱼内脏的油脂提取提供了一定的理论指导。
Squid viscera contains abundant crude fat and protein.The crude oil take up about 20 ~ 30 percent of squid viscera(dry weight),unsaturated fatty acids accounted for about 65%,EPA and DHA accounted for about 32%of total fatty acids,so squid viscera is a good raw material for acquiring EPA and DHA.In this paper,we used different methods to extract squid viscera oil,the extraction rate was calculated and fatty acid composition was analysed using GC-MS.Organic solvents extraction,enzymatic hydrolysis,alkaline hydrolysis were used to obtain the squid visceral oil to compare the extraction rate,Organic solvent extraction method was finally selected.Several kinds of organic solvent were used such as chloroform-methanol,ethanol and n-hexane.The average extraction rate of three methods can reach 60.61%,17.71%and 22.72%,respectively.At the same time,the fatty acid composition analysis of oil extracted by different methods was carried out.The results showed that the average content of the unsaturated fatty acids was 26.24%,and the average content of polyunsaturated fatty acids was39.01%.In polyunsaturated fatty acids,the content of EPA was 7.14%,and the content of DHA was 24.97%.The fatty acid content of squid visceral fat is rich,and its composition is similar to that of fish oil.Different solvent extraction has no significant effect on the fatty acid content of squid viscera oil,but it has some influence on the type of fatty acid isomers that is extracted.The most types of fatty acids were extracted by hexane,followed by enzyme and hexane,chloroform methanol method,the least ethanol method.The results of this paper provide theoretical guidance for extraction of the oil from squid viscera.
