藏猪源益生芽孢杆菌的筛选鉴定及部分益生特性研究

摘要
从藏猪粪便中分离筛选出具有良好益生潜力的益生芽孢杆菌,为藏猪源益生菌的开发奠定基础。先将健康藏猪粪便滤液于90℃水浴处理10分钟杀死样品中的非芽孢杆菌,涂布于培养基后挑取单菌落纯化并扩大培养,再进行拮抗试验筛选能抑制常见病原菌的芽孢杆菌,综合形态学、生理生化特征及16S rDNA序列分析对筛选菌株进行鉴定,通过菌株耐受性试验、产水解酶试验、抗生素敏感试验等对菌株生物学特性进行研究。筛选到3株对常见病原菌具有一定抑制作用的芽孢杆菌,其中解淀粉芽孢杆菌SWUN-TP23在不利环境中具有较强的稳定性,可代谢产生纤维素酶、淀粉酶和蛋白酶,菌体内不含抗常用兽用抗生素的耐药基因。分离筛选到的解淀粉芽孢杆菌SWUN-TP23有良好的潜在益生能力,具有进一步开发研究的价值。
Probiotic bacillus with good potential,which were isolated and screened from Tibet Pig feces,provide a basis for the development of probiotics of Tibet Pig source.Firstly,the healthy Tibetan Pig feces filtrate adopted the 90 ℃water-curing treatment for 10 minutes to kill non-bacillus coli in samples,after being coated on culture media,the single colonies were picked to purify and expanded,and then screened Bacillus capable of inhibiting common pathogenic bacteria by antagonistic tests.Through morphological,physiological and biochemical characteristics and 16 S rDNA sequence analysis,the selected strains were identified.Through tolerance tests,enzyme production tests and antibiotic sensitivity tests,the biological characteristics of strains were studied to select the best strains.Through screening,3 strains of Bacillus have a certain inhibitory effect on the common pathogenic bacteria.Among them,Bacillus subtilis SWUN-TP23 has a strong stability in the adverse environment.It has strong stability in adverse environment and can produce cellulase,amylase and protease.Bacillus amyloliquefaciens SWUN-TP23 does not contain drug resistance genes against commonly used veterinary antibiotics.Bacillus amyloliquefaciens SWUN-TP23,through isolation and screening,has good potential probiotic ability and is worthy of further studying and developing.
