摘要
2,3-丁二醇是一种极具价值的液体燃料,燃烧值为27k J/g。由于化学合成成本高、过程繁琐、不易操作,因而微生物法生产2,3-丁二醇成为研究热点。本研究旨在利用UV诱变提高产酸克雷伯氏菌的2,3-丁二醇产量。首先对产酸克雷伯氏菌进行UV诱变的条件筛选,筛选照射高度、照射时间及菌悬液浓度等条件,得出最佳诱变条件为照射高度15cm、照射时间30s、菌悬液浓度为10~6CFU/ml。接着在最适条件下对产酸克雷伯氏菌进行紫外诱变筛选出高产2,3-丁二醇的菌株,并且通过多代连续培养观测其遗传稳定性,筛选不退化的菌株保存。
2, 3-butanediol is a valuable liquid fuel and has a combustion value of 27 k J/g.Since the high cost,complicated process and difficult operation of Chemical synthesis, microbial production of 2, 3-butanediol has become a research hotspot. This study aims to utilize UV mutagenesis to improve the 2, 3-butanediol yield of Klebsiella oxytoca.Firstly,conduct the condition screening of UV mutagenesis to search the optimal exposure height 15 cm,irradiation time 30 s and Bacteria suspension concentration 10~7 CFU/ml. Then mutate the Klebsiella oxytoca under the optimal condition to screen the strain which can highly produce 2,3-butanediol.Afterwards,observe the hereditary stability of mutated strains through continous culture of many generations,so as to get unabated strain.
