降低琼脂中硫酸根含量的研究

摘要
硫琼脂中的硫酸基团会降低琼脂的凝胶强度,所以在商业生产琼胶过程中需要去除。本实验以江蓠琼脂为原料,采用阴离子交换树脂法、碱法和EDTA-Na_2法处理琼脂,研究这些方法对琼脂硫酸基团脱除的能力。结果表明,用D296和D318阴离子交换树脂不能脱除琼脂中硫酸根的含量;碱法处理琼脂时,60℃条件下琼脂中硫酸根含量低于80℃,且得率高于80℃;用EDTA-Na_2法脱除琼脂硫酸根时,其脱除琼脂粉中硫酸根的效果要优于脱除琼脂溶液及凝胶时的效果。采用阳离子交换树脂、碱和EDTA-Na_2两两结合处理琼脂发现碱与阳离子交换树脂联合处理后得到的琼脂产品中硫酸根的含量最低,为0.08%。采用碱、阳离子交换树脂和EDTA-Na2联合处理琼脂,硫酸根为0.12%。
Because sulfate groups in sulfur agar reduce the gel strength of agar,they needed to be removed in the process of commercial agar production.Thus,anion-exchange resin,alkali and EDTA-Na_2 were used to reduce the content of sulfate in this study.It was shown that agar samples seperately treated with D196 and D318 resins made no significant difference in sulfate content.Yet,agar treated by alkaline at 60℃ had lower sulfate content and higher yieldthan the sampledid at 80℃.Agar powdertreatedwith EDTA-Na_2 exhibited a better ability to reduce sulfate content than the agar sol and agar gel treated with EDTA-Na_2,respectively.Then,agar samples were treated by the different combination methods of cation-exchange resin,alkaline and EDTA-Na_2.As a consequence,it showed the lowest sulfate content(0.08%) in the agar when assembly treated by alkaline and cation-exchange resin.Besides,cation-exchange resins combined with alkaline and EDTA-Na_2 Were also used to treat agar,in which the content of sulfate were 0.12%.
