激发发射矩阵荧光结合二阶校正方法同时测定人体血浆中的华法林和阿司匹林含量

摘要
华法林和阿司匹林两种药物联用既可以用于深静脉血栓、肺栓塞等血栓性疾病的治疗,又克服了华法林治疗范围窄以及阿司匹林因剂量过大而产生各种副作用的缺点。但这两种药物的联用也大大增加了出血的风险,因此,测定它们的血药浓度可以为临床合理联用药物提供依据。已经有研究采用高效液相色谱联用紫外检测器(HPLC-UV)对复方片剂或人体血浆中的华法林和阿司匹林进行同时定量分析,这种方法耗时费力,并会消耗大量溶剂。华法林和阿司匹林的荧光光谱重叠严重,并且人体血浆的背景荧光干扰较强,传统的荧光分析方法无法对其进行直接测定。本文采用激发发射矩阵荧光结合基于交替惩罚三线性分解(APTLD)算法的二阶校正方法对人体血浆中的华法林和阿司匹林进行同时定量分析,当选取的组分数为4时,得到华法林和阿司匹林的平均回收率分别为109.0%和99.53%,相关系数均大于0.99。该方法能够简单、快速地检测人体血浆中的华法林和阿司匹林含量,对临床用药的监测和指导具有非常重要的意义。
The combination of warfarin and asprin can be used for the treatment of thrombotic diseases such as deep vein thrombosis and pulmonary embolism,overcoming the disadvantages of narrow scope therapy of warfarin and various kinds of side effects of aspirin owing to its excessive doses,which also greatly increased the risk of bleeding.In this paper,a novel method that combines excitation-emission matrix fluorescence with second-order calibration method based on alternating penalty trilinear decomposition(APTLD) algorithm was developed to simultaneously and directly determinate the warfarin and asprin contents in human plasma even in the presence of matrix interferences.When the component number was chosen 4,the obtained average recoveries were 109.0%for warfarin,and 99.53%for asprin,respectively,and the correlation coefficient of both ones were more than 0.99.Therefore,the proposed method is of great significance to the monitoring of clinical administration and also being simple sample pretreatment and low cost.
