富硒滑子蘑菌丝体和子实体中半胱氨酸及其合成关键酶的研究

半胱氨酸在生物体内具有多种生理功能,也是合成谷胱甘肽的重要物质。本论文研究了不同富硒浓度对滑子蘑的菌丝体和子实体中半胱氨酸及半胱氨酸合成酶(CSase)的影响,培养浓度为:0、20、40、60、80、100mg/kg。采用强酸性茚三酮检测酶促反应,通过亚硝酰铁氰化纳法测定半胱氨酸含量。实验结果:菌丝体和子实体中的CSase活性及半胱氨酸含量随着富硒浓度的增加整体呈先上升在下降的曲线,但它们的半胱氨酸含量及其合成关键酶都大于相对应的空白组;子实体中的CSase活性略大于菌丝体中的CSase活性,富硒浓度为60mg/kg时,菌丝体的CSase具有最大活性为0.047 U/mmol;富硒浓度为40mg/kg时,子实体的CSase具有最大活性为0.076U/mmol;子实体中的半胱氨酸含量大体上和菌丝体中的半胱氨酸含量相等且都大于对应的空白组,在富硒浓度为80mg/kg时菌丝体中的半胱氨酸含量急剧上升,达到408.6μg/mg;菌丝体中的半胱氨酸含量在富硒浓度为60mg/kg达到峰值222.8μg/mg。进而得到结论,富硒对滑子蘑生长过程中半胱氨酸的合成有一定的促进作用。
Cysteine in organism,has many physiological functions,but also an important material of synthesis of glutathione.This paper studied the effect of different concentration of Se-enriched Pholita Namek mycelium and fruiting body of its cysteine and cysteine synthase(CSase),the training concentration as:0,20,40,60,80,100 mg/kg.Using a strongly acidic ninhydrin method detected enzymatic reaction,the content of cysteine was measured by sodium nitroprusside method.The results:CSase activity and the content of cysteine in mycelium and fruiting body were in a curve that with the increase of selenium concentration it first rised then declined,and each of them were greater than the corresponding blank group;CSase activity in fruiting body was slightly larger than that of mycelium,in the Se-enriched concentration of 60 mg/kg,CSase activity in mycelium having a maximum value,0.047 U/mmol;in the Se-enriched concentration of 40 mg/kg,CSase activity in fruiting body having a maximum value,0.076 U/mmol;Cysteine content in fruiting body and mycelium were generally equal and both of them were larger than the corresponding blank group,in the Se-enriched concentration of 80 mg/kg,the content of cysteine in the mycelium rose sharply,reaching 408.6 μg/mg;cysteine content in mycelium had a peak at Se-enriched concentration was 60 mg/kg,222.8 μg/mg.Then come to a conclusion,Se-enriched on Pholita Namek growth process could have certain role in promoting about cysteine synthesis.
