齐口裂腹鱼脾脏转录组差异基因分析

摘要
为探讨齐口裂腹鱼在温和气单胞菌感染后的免疫应答机制,采用RNA-Seq技术对齐口裂腹鱼感染温和气单胞菌(0 h、24 h和48 h)脾脏组织进行转录组测序。将Unigene在NR、GO和KEGG数据库注释,并对不同组差异基因表达进行分析。结果显示:脾脏Unigene总数为97,581,在Nr数据库中成功注释27,887条,在GO数据库中成功注释25,105条,在KEGG数据库中成功注释11,884条。24 h与0 h之间差异基因数为1044个,差异基因富集的122条代谢通路中有7条为显著性富集通路。48 h与0 h相比,差异基因数为1024个,差异基因富集的128条代谢通路中有9条为显著性富集通路。48 h和24 h相比,差异基因数目为886个,差异基因富集的104条代谢通路中有4条为显著性富集通路。三个组共有信号通路为NF-κB的信号通路和低氧诱导因子-1信号通路,并从富集通路中获得了大量免疫相关基因。这为了解齐口裂腹鱼抗菌免疫机制提供参考。
To characterize the immune mechanism of Schizothorax prenanti(S. Prenanti) infected with Aeromomas sobria(0 h, 24 h, 48 h). Splenic transcriptomes of Aeromomas sobria infected S.Prenanti(normal 0 h, treated 24 h, treated 48 h) were analyzed by Illumina RNA-seq. Unigenes were annotated by NR, GO and KEGG database, and genes of differentially expressed in different groups were analyzed respectively. The results showed that 97,581 unigenes were obtained from raw datum of three groups. There were 27,887 unigenes successfully annotated in Nr database,25,105 unigenes in GO, and 11,884 unigenes in KEGG. 1044 differential genes were found by transcriptomes(24 h VS 0 h), which had 7 enrichment pathways of significance in 122 metabolic pathways. 1024 differential genes were obtained by transcriptomes(48 VS 0 h), which had 9enrichment pathways of significance in 128 metabolic pathways. 886 differential genes were found by transcriptomes(48 h VS 24 h), which had 4 enrichment pathways of significance in 104 metabolic pathways. There were two same signaling pathways, including NF-kappa B signaling pathway and HIF-1 signaling pathway. Lots of the immune-related genes were selected and analyzed from the enrichment pathways. It will benefit the further study of immune regulation of Schizothorax prenanti.
