刺参自溶过程中体腔细胞Ca~(2+)信号与其凋亡相关性研究

本研究考察刺参(Stichopus japonicus)自溶过程中,其体腔细胞Ca~(2+)信号与凋亡的相关性。将105只刺参随机等分为处理组、抑制组和对照组。处理组刺参经UVA(15W/m~2)照射30min,室温避光静置0、1、2、3和4h,制备体腔细胞;抑制组刺参经Ca~(2+)螯合剂(BAPTA-Na,IC_(50)=1.47mM)灌入体腔内,UVA照射、静置后制备体腔细胞;对照组刺参未经UVA照射,制备体腔细胞。应用流式细胞仪检测体腔细胞内Ca~(2+)浓度([Ca~(2+)]_i)和细胞凋亡率;应用酶标仪检测Ca~(2+)信号相关酶(Ca~(2+)-ATP酶、钙调神经磷酸酶和钙蛋白酶)的酶活水平。结果表明,与对照组相比,处理组体腔细胞[Ca~(2+)]_i随着静置时间的延长而逐渐升高,静置2h时达到最高值,随后下降;Ca~(2+)信号相关酶活变化与[Ca~(2+)]_i变化趋势相一致;细胞凋亡率静置3h最高,随后降低。然而,抑制组体腔细胞[Ca~(2+)]_i、SERCA酶活和细胞凋亡率,虽与处理组的变化趋势相一致,但其表达量均显著低于处理组。综上结果揭示,UVA诱导刺参自溶过程中,其体腔细胞Ca~(2+)发生稳态失调,激活Ca~(2+)信号相关酶活表达,进而导致体腔细胞凋亡。
In this study,the relationship between calcium signals and coelomocytes apoptosis during sea cucumber(Stichopus japonicus) autolysis was investigated.Stichopus japonicus(n = 105) were randomly and equally dividedinto treatment group,Ca~(2+)-suppression group and control group,respectively.The treatment group wasirradiated by UVA with the intensity of 15w/m~2 for 30 min and incubated at room temperaturein dark for 0,1,2,3,and 4h,respectively,and then the coelomocytes were prepared.The Ca~(2+)-suppression group was injected with calcium chelation agent(BAPTA sodium salt,IC_(50) = 1.47mM)into the coelomic cavity and the coelomocytes were collected after UVA irradiation and incubation.The control group was treated without UVA irradiation.The intracellular free calcium concentration([Ca~(2+)]_i) and apoptotic rates of coelomocytes were detected by Flow cytometer.Calcium signals-related enzymes(sarcoplasmic-endoplasmic reticulum Ca~(2+)-ATPase,calcineurin and calpain) wereexamined by microplate reader.The Results indicated that,comparing with control group,[Ca~(2+)]_i of coelomocytes increased gradually with the extended incubation time toreach the highest level at 2h and then decreased.Activity changes of calcium signals-related enzymes demonstrated a similar trend to change of Ca~(2+) signals.Apoptosis rates of coelomocytes achievedthe highest value at 3h and then declined.Although the changes of[Ca~(2+)]_i SERCA activity and apoptosis rates in Ca~(2+)-suppression group were agreed with that of the treatment group,the expression of these parameters were markedly lower compared to treatment group.Our findings revealed that,the unbalancedintracellular Ca~(2+) homeostasis and activated calcium signals-related enzymess might be involved in coelomocytes apoptosis during UVA-induced autolysis of Stichopus japonicus.
