鱼类干扰素调节因子4(IRF4)基因的表达:同源基因功能存在分化

在哺乳动物中,干扰素调节因子4(IRF4)在B细胞、T细胞和树突状细胞的发育和分化中发挥重要作用。同时,IRF4能调控IRF5、My D88、IL21、PGC1?和NOD2的表达。本研究首次研究了IRF4同源基因(IRF4a、IRF4b)以及它们相关基因的表达规律。结果显示:IRF5在肌肉中含量丰富,其余基因均为免疫相关组织中出现高表达。IRF4b在斑马鱼胚胎发育中呈现母源表达,而IRF4a在出膜前表达水平高,这说明它们在斑马鱼先天免疫中发挥重要作用。草鱼呼肠孤病毒和鲤春病毒均能诱导IRF4a和IRF4b的表达,且两者表达规律截然不同,这些说明两者功能发生了分化。
In mammals, interferon regulatory factor 4(IRF4) plays an important role in the process of development and differentiation of B cells, T cells and dendritic cells. It can regulate immune pathway through IRF5, My D88, IL21, PGC1?, and NOD2. In the present study, we investigated the expression pattern of IRF4 paralogues and these related genes for the first time in teleosts. The results showed that these genes were all expressed predominantly in known immune tissues while IRF5 was also relatively highly expressed in muscle. IRF4 b showed maternal expression in the oocyte and the higher expression of IRF4 a before hatching might be involved in the embryonic innate defense system. GCRV and SVCV can induce the expression of IRF4 a and IRF4 b in zebrafish. Furthermore, IRF4 a and IRF4 b displayed a distinguishing expression pattern in vivo and in vitro, suggesting that IRF4 paralogues might play different roles in immune system.
