新型苯并噻唑衍生物作为pH荧光探针应用于细胞成像

摘要
细胞内pH对于调控许多细胞的生理过程发挥重要的作用~([1]),pH值异常往往导致细胞功能紊乱~([2])。因此,对细胞内pH变化进行灵敏检测,有利于从分子水平上研究细胞的生理病理过程。本文利用乙烯基桥连苯并噻唑(电子给体,D)和喹啉醛(电子受体,A)设计合成了基于分子内电荷转移(ICT)效应的pH荧光探针(BTVQ)。以317 nm为激发波长,BTVQ的最大荧光发射位于428 nm(pH=7),随着pH由7.0降低到2.2,该发射峰逐渐减弱(Fig.1a)。通过激光共聚焦荧光成像技术,BTVQ成功实现人子宫颈鳞癌Siha细胞中pH变化的高灵敏响应(Fig.1b),可作为新型pH探针应用活细胞内酸性环境pH变化的检测。
Intracellular p H play an important role for regulating many cellular processes~([1]). The monitoring of p H changes in living cells, is critical for studying cellular functions and further understanding physiological and pathological processes. A novel p H fluorescent probes(BTVQ) based on intramolecular charge transfer(ICT) process was synthesized via the ethylene bridging of benzothiazole(D) and 2-quinolinecarbaldehyde(A). BTVQ exhibited fluorescent emission at 428 nm with an excitation wavelength of 318 nm(p H=7). Upon decreasing the p H from 7.0 to 2.2, the fluorescence intensity reduces gradually(Fig. 1a). The confocal fluorescent microscopic images of the highly sensitive p H response in Siha cells were achieved successfully(Fig. 1b), suggesting that BTVQ could be used as a novel p H probe for effectively detecting pH changes in acidic organelles.
