大孔径树脂纯化油茶多酚工艺条件研究

摘要
为利用油茶有效成分多酚类化合物,采用大孔径树脂纯化油茶多酚并测定其体外抗氧化活性。通过静态吸附试验和动态吸附单因素实验等确定纯化大孔径树脂类型。通过研究上样浓度、上样量、pH值、溶剂流速等影响因素获得最佳纯化条件,通过DPPH法测定油茶多酚体外抗氧化活性实验。结果HPD400是最佳油茶多酚纯化树脂,最佳纯化条件为:上样浓度为2.5 ug/mL,上样量为4BV,上样pH为2,解吸液浓度为50%乙醇。在此条件下纯化后多酚含量为34.93mg/g,与原多酚提取液相比多酚含量提高了52.4%,且油茶多酚对DPPH自由基有一定的清除能力。
To make full utilize of polyphenols from camellia,purifying polyphenol by macroporous resinand studying its antioxidative property in vitro were carried out.The best type of the resin was selected through the static adsorption test,static adsorption dynamic curve and dynamic adsorption experiment,etc.Parameters studied affected purifying factors including sample concentration,loading quantity of sample,pH value of sample,and flow rate of solvent,etc.Vitro antioxidative property of camellia polyphenol was assessed by DPPH radical ability.Studis showed that HPD400 was the best type of purifying resin.The optimum conditions for purification camellia polyphenol were as follows:2.5 ug/mL sample concentration,4 BV samplevolume,sample pH at 2,and 50%ethanol resolution.Polyphenolscontent of the extraction was 34.93mg/g after purification,which is 52.4%higher than the original extract.DPPH radical scavenging ability of camellia polyphenol was also high.
