响应面法优化菌草灵芝多肽螯合硒离子的研究

摘要
目的:为优化菌草灵芝多肽-硒螯合物的制备工艺,在单因素的基础上应用响应面法优化了菌草灵芝多肽-硒螯合物的制备条件,为合成有机硒化合物提供了一种新的原料。方法:利用复合蛋白酶酶解菌草灵芝蛋白,以亚硒酸钠为硒源与菌草灵芝蛋白酶解物进行螯合,对亚硒酸钠溶液与蛋白酶解液的体积比、反应温度、反应时间和pH的交互作用进行试验探讨并确定了最优水平。结果:最佳的螯合工艺条件为时间60min,温度75℃,pH9,亚硒酸钠溶液与酶解液的体积比为1:2。该条件下,螯合物中硒含量为2985.89±10.59μg/g。由红外光谱分析可知,菌草灵芝多肽和硒离子发生螯合反应,生成了一种新型的螯合物。讨论:经过复合蛋白酶酶解的菌草灵芝多肽能够与硒离子螯合形成菌草灵芝多肽-硒螯合物,为菌草灵芝的开发利用提供了一个新的思路。
Objective:To optimize the preparation technology of JUNCAO Ganoderma lucidum peptide-selenium chelate and provide a new source for organic seleniun compounds,the response surface method based on single factor tests was used to study the chelation conditions.Methods:The proteins from JUNCAO Ganoderma lucidum were hydrolysed by compound protease,and then the peptides were chelated with sodium selenite,which was a kind of inorganic selenium.The interaction effects of volume ratio of sodium selenite solution to JUNCAO Ganoderma lucidum protein enzymatic hydrolysate,reaction time,temperature and pH were investigated and discussed to determine their optimum levels.Results:The optimal chelating conditions were volume ratio 1:2(v:v),reaction time 60 min,reaction temperature 75 t,and pH 9.0.Under the optimal reaction conditions above,the content of selenium in the chelate was 2985.89 ± 10.59 μg/g.The infrared spectral analysis revealed that the chelation reaction between JUNCAO Ganoderma lucidum peptides and selenium ions happened,and generated a new-style chelate.Conclusion:The peptides produced by JUNCAO Ganoderma lucidum protein enzymatic hydrolysis could chelate with selenium ions to produce a new kind of chelate.This paper provided a new approach of development and utilization of JUNCAO Ganoderma lucidum.
