马氏珠母贝组蛋白H4-1和H4-2基因的克隆与表达分析

摘要
组蛋白(histone)是真核细胞中与DNA结合用来形成染色质的一类特殊蛋白,参与了DNA的复制、损伤修复和转录等过程。为了探究马氏珠母贝组蛋白H4的序列及表达特征,本文运用cDNA末端快速扩增(RACE)技术克隆得到马氏珠母贝H4(Pinctada martensii H4,PmH4)的两种表达基因,分别为PmH4-1和PmH4-2。SMART软件分析显示PmH4-1和PmH4-2均具有典型的H4结构域。多序列比对结果表明,PmH4-1与其它物种的H4氨基酸序列相似度较高,在98.06%-100.00%之间,而PmH4-2与其它物种的H4氨基酸序列相似度较低,在53.40%-55.88%之间;PmH4-1与PmH4-2间的氨基酸序列相似度也较低,仅为55.34%,说明马氏珠母贝在进化过程中除了常规的组蛋白H4,可能进化出特殊的组蛋白H4。荧光定量显示,PmH4-1和PmH4-2基因在马氏珠母贝外套膜边缘区和中央区、鳃、血细胞、闭壳肌、足、性腺、肝胰腺等7种组织中均有表达,其中,PmH4-1在性腺中高表达,PmH4-2在外套膜的边缘膜中高表达,PmH4-1在各组织中表达量均显著高于PmH4-2(P<0.01)。LPS刺激后,马氏珠母贝血细胞中PmH4-1基因的相对表达量变化不显著(P>0.05),而PmH4-2基因的相对表达量上升,并在12 h后达到峰值,随后下降到正常水平(P<0.05),说明PmH4-2可能与马氏珠母贝的免疫反应有关,而PmH4-1可能是一种常规组蛋白,不参与机体的免疫反应。本研究可为进一步探讨PmH4及其变体在马氏珠母贝中的免疫防御机制和生物学特性提供理论依据.
Gonadotropin-releasing hormone(GnRH) plays important roles in reproduction. In the present study, we demonstrated the existence of GnRH-like peptides in the central nervous system(CNS) and In order to explore the sequence and expressive characteristics of histone H4 in Pinctada martensii,using Rapid Amplificationof cDNA End(RACE) Cloning technology, we have got two full length sequence of histone H4 cDNA from P. martensii(PmH4-1 and PmH4-2).SMART software analysis indicated that both of PmH4-1 and PmH4-2 had a typical H4 structure domain. Multiple sequence alignment showed PmH4-1 was highly conserved among species, the sequence identities ranged between 98.06% and 100.00%. While PmH4-2 was lowly conservative compared with H4 from other species, the sequence identities ranged between 53.40% and55.88%.
