MC-LR对凡纳滨对虾相关免疫酶活力及基因表达量的影响

本实验通过设置对照组和不同浓度梯度的微囊藻毒素(MC-LR)实验组(分别为0;0.50;0.72;1.03;1.47;2.11;3.00μg/L MC-LR)探究MC-LR对凡纳滨对虾(Litopenaeus vannamei)肝脏免疫相关酶活力及其基因表达量的影响。结果表明,在30d后发现MC-LR能显著影响凡纳滨对虾肝胰脏的免疫相关酶活力及基因表达量(P<0.05)。SOD、GPx和POD活力均随着毒素浓度的增加而逐渐增大,毒素浓度为3.0μg/L时达到最大值,分别为250.40 U/mg、125.11 U/mg和7.42 U/mg。ACP和AKP活力呈先增加后减小的变化趋势,当毒素浓度为0.72μg/L时达到最大值,分别为76.97 U/mg和91.56 U/mg。当毒素浓度大于0.50μg/L时,LZM活力随着毒素浓度的增加而呈现下降的趋势,毒素浓度为1.47μg/L时达到最小值,为9.69U/mg。随着毒素浓度的增加,c Mn-sod和gpx基因表达水平显著上调,lzm基因表达水平显著下调,与相应的酶活力变化相一致。表明水体中长期的低浓度的MC-LR能引起凡纳滨对虾的应激反应,促使对虾对免疫酶及相关基因进行调节以应对毒素的刺激。
The study was conducted to investigate the effects of microcystins(MC-LR) on the activity and gene expression of immune-related enzymes in the hepatopancreas of white shrimp Litopenaeus vannamei by treating with different concentrations of MC-LR(0; 0.50; 0.72; 1.03;1.47; 2.11; 3.00 μg/L MC-LR). The results showed that the activities and gene expression of immune-related enzymes in the hepatopancreas of L. vannamei were significantly affected by MC-LR after 30 days(P<0.05). With the increasing concentration of WC-LR, the activities of SOD 、 GPx and POD reached their peak values at concentration of 3.0μg/L, 250.40U/mg 、125.11U/mg and 7.42U/mg, reapectively. ACP and AKP activities inereased at the concentration of 0.72μg/L and decreased after that. The peak values were 76.97 U/mg and 91.56 U/mg. LZM activity reached its minimum value at concentration of 1.47μg/L with the increasing concentration of WC-LR, The minimum values was 9.69 U/mg. With the increasing concentration of WC-LR,the gene expression level of c Mn-sod and gpx up-regulated while the gene expression level of lzm down-regulated significantly, which were consistant with the variation of their corresponding enzymes.
