碱-氧结合法制备牛毛角蛋白的研究

摘要
近几年来,随着保毛脱毛法清洁生产工艺的逐步实施,如何处置越来越多的废弃牛毛成为亟待解决的新课题。本文采取碱-氧结合法对保毛脱毛所得牛毛水解提取角蛋白,以NaOH浓度,H2O2浓度,水解温度,搅拌速率,设计4因素正交实验,获得提取牛毛角蛋白的最佳条件:水解温度82℃,固液比1:20,H_2O_2浓度15%,NaOH浓度1.5 mol/L,水解时间1 h,搅拌速率200 r/min。研究进一步考察了单因素实验下水解液的粘度和角蛋白得率,发现了最优条件下所得的水解液粘度为0.8349 cp,角蛋白得率达到27.24%。红外光谱(FT-IR)和热重分析(TGA)表明水解过程中,牛毛中的双硫键、肽键发生了部分断裂,导致提取后的角蛋白热稳定性弱于牛毛的热稳定性。
In recent years,with the gradual implementation of clean production process for hair-preserving unhairing technology,the reuse of waste hair was an urgently problem needs to be solved.In this paper,the method of alkali oxidation method was used to extract the keratin from the bovine hair.With the concentration of sodium hydroxide,hydrogen peroxide concentration,hydrolysis temperature,the stirring speed of the mixer as the variable value,the orthogonal experiment was designed.The experimental results indicated that the optimum reaction conditions of alkali oxidation binding method were as following:82 ℃,solid-liquid ratio of 1:20,15%H_2O_2,1.5 mol/L NaOH hydrolysis for 1h,stirring speed 200 r/min.On this basis,the paper further investigated the viscosity of the hydrolysate and keratin yield.The results indicated that under the optimal conditions,the viscosity of hydrolysate was 0.8349 cp,the keratin yield reached 27.24%.The FT-IR and TGA analysis showed that the bovine hair disulfide bond and peptide bond were ruptured in the process of hydrolysis.The thermal stability of bovine hair was better than bovine hair keratin.
