PLGA多功能纳米诊疗剂的制备及其在肿瘤可视化光热治疗中的应用

摘要
纳米诊疗剂是通过纳米合成技术制备出具备诊断和治疗双重特性的多功能纳米药物载体。本实验采用配位组装技术,制备多功能聚乳酸羟基乙酸(PLGA)纳米诊疗剂,并进行核磁成像引导下的肿瘤光热治疗。首先采用复乳法制备PLGA空白纳米粒,其次在其表面通过多巴胺的自聚反应修饰一层薄层(PDA),最后通过配位组装将Mn离子结合至PDA@PLGA的表面,构建Mn~(2+)-PDA@PLGA纳米诊疗剂。实验结果表明,在808nm红外光照射下,PDA良好的光热转换性能,可显著升高体系温度,到达光热治疗目的,同时,Mn离子作为临床分子影像造影剂对疾病进行诊断,实现肿瘤的可视化治疗。
Nanotheranostics is multifunctional nano drug carriers prepared by nano-synthesis technology with dual diagnosis and treatment characteristics. In this paper, the PLGA microcapsules were generated by the water-in-oil-in-water(W/O/W) double emulsion method, which are then modified with polydopamine(PDA) through the self-polymerization of dopamine. Finally, the Mn~(2+)-chelated PDA@PLGA nanoparticles were obtained as novel theranostic agents for simultaneous MRI signal enhancement and photothermal ablation of cancer cells. Therefore, the developed multifunctional PLGA nanoparticles could be used as a promising theranostic agent for MRI-guided photothermal therapy of cancer cells.
