不同时期冷藏金枪鱼优势腐败菌多样性及生物胺变化规律的研究

摘要
研究了不同时期冷藏金枪鱼优势腐败菌多样性及生物胺的变化规律。通过生理生化鉴定、16S rRNA序列分析鉴定及系统发育树的建立,以及生物胺(腐胺Put、组胺His、酪胺Tyr)的检测,分析得到4℃冷藏8天内的金枪鱼菌优势腐败菌多样性及生物胺的变化规律,并最终分离筛选到21株不同特征的纯化菌株。结果表明,随着贮藏时间的延长,金枪鱼的生物胺含量逐渐升高,其中组胺含量最高,而酪胺含量始终处于低水平状态;冷藏金枪鱼初始菌相主要构成为嗜油不动杆菌(Acinetobacter oleivorans)、莓实假单胞菌(Pseudomonas fragi)、热死环丝菌(Brochothrix thermosphacta)、黄歇假单胞菌(Pseudomonas fulva);微生物菌落结构在贮藏期间不断变化,多样性降低,在货架期第8天逐渐形成以不动杆菌(Acinetobacter spp.)、假单胞菌(Pseudomonas spp.)为主的微生物群落结构。
The changes of dominant microflora and biogenic amines of tuna(Thunnus obesus) during chilled storage were studied by henotypic analysis 16 S rRNA gene sequencing analysis and the establishment of phylogenetic tree,and the content of biogenic amines(putrescine,histamine,tyramine) changes of tuna were researched regularly during iced storage.Finally,21 bacteria strains of tuna stored under 4℃ were obtained.The results showed that the content of biogenic amines were increased with the extension of storage time,and histamine content were increased rapidly and became the major biogenic amines,while tyramine was low in all storage time.The initial microbial flora was dominated by Acinetobacter oleivorans,Pseudomonas fragi,Brochothrix thermosphacta,Pseudomonas fulva.The structure of microbial growth kept changing during storage,and the bacterial diversity of tuna decreased along with storage time.On the shelf-life of eighth day Acinetobacter spp.,Pseudomonas spp.,Brochothrix thermosphacta became the dominant bacteria in the microbial community.
