日粮标准回肠可消化缬氨酸与赖氨酸比对仔猪采食量调控的影响机制

本研究通过两个试验探讨日粮补充缬氨酸(valine,Val)对仔猪采食量的影响。试验一研究日粮补充合成Val对仔猪采食量、血浆氨基酸和采食调节因子的影响。18头初始体重为(18.9±2.3)kg的杜×长×大三元杂交仔猪(公母各半)随机分为3个处理,每个处理6个重复。对照组基础日粮中SID Val与Lys的比例为0.45,两个处理组日粮SID Val与Lys的比分别为0.65和0.85。试验期为14天。结果表明,随着日粮中SID Val:Lys比例的提高,仔猪平均日采食量、平均日增重和增重耗料比线性(P<0.01)和二次(P≤0.05)提高;血浆中Val的水平随日粮中SID Val:Lys提高线性增加(P<0.01);相比对照组,日粮SID Val:Lys为0.85时可显著降低胃底腺黏膜胆囊收缩素(cholecystokinin,CCK)的mRNA表达量(P<0.05),日粮SID Val:Lys为0.65时,可显著降低下丘脑阿黑皮素原(Proopiomelanocortin,POMC)的mRNA表达量(P<0.05)。试验二探究日粮补充合成Val对仔猪餐后血浆氨基酸含量的影响。6头初始体重为(15.7±0.8)kg杜×长×大三元杂交仔猪(公母各半)随机分为3个处理,每个处理2个重复。日粮配方组成同试验一。仔猪单栏饲养在代谢笼,在前腔静脉进行血插管手术。结果表明,餐后1、2、3和4 h,仔猪采食日粮SID Val:Lys为0.65和0.85时的血浆Val水平显著高于(P<0.05)采食日粮SID Val:Lys为0.45时。总之,日粮添加Val能够显著提高仔猪的采食量,进而提高生长性能,其可能的机制是日粮中Val水平影响仔猪餐后血浆Val水平,进而降低了胃底腺黏膜CCK和下丘脑POMC的表达量来调控采食量。
Two experiments were conducted to study the effect of supplemental crystalline valine(Val) on feed intake of piglets.In Exp.I,18 pigs,with an initial body weight of18.9 ± 2.3 kg,were divided into 3 treatments with 6 replicates per treatment to study the effect of standardized ileal digestible(SID) Val to lysine(Lys) ratios on feed intake and the expression of endocrine regulatory factors of piglets.The basal diet(control treatment) was formulated to contain SID ratio of 0.45,and crystalline Val was added to the basal diets to form SID Val:Lys ratios of 0.65 and 0.85,respectively.The experiment lasted for 14 d.Average daily feed intake(ADFI),average daily gain(ADG) and gain to feed ratio(G:F) increased(linear and quadratic,P≤ 0.05) with the increase of dietary SID Val:Lys ratio.Plasma Val content increased linearly(P<0.01) with more supplemental Val.Compared with the control treatment,the expression of gastralcholecystokinin(CCK) was lower(P<0.05) in pigs fed diet of SID Val:Lys ratio of 0.85.The expression of hypothalamicproopiomelanocortin(POMC) was lower(P<0.05) in pigs fed diet of SID Val:Lys ratio of 0.65 than the control treatment.In Exp.Ⅱ,6 pigs,with an initial body weight of 15.7 ± 0.8 kg,were divided into 3 treatments with 2 replicates per treatment to study the effect of dietary SID Val:Lys ratios on the postprandial plasma AA concentrations of pigs.The diets were the same as in Exp.Ⅰ.Pigs were raised individually in metabolism crates,and cathetered in the precaval vein.Results showed that postprandial plasma Val concentration was higher(P<0.05) in pigs fed diets of SID Val:Lys ratios of 0.65 and 0.85 than the control treatment at 1,2,3 or 4 h after meal.In conclusion,diets supplemented with crystalline Val increased the feed intake and growth performance of piglets,and the regulatory mechanisms in feed intake may be that dietary crystalline Val increased the plasma Val content and decreased the CCK and POMC mRNA expression.
