酶解海参内脏制备ACE抑制肽的研究

摘要
海参内脏具有极其重要的食用价值,但在我国却作为海参加工的副产物而未被充分利用。本文以海参内脏为原料,选用中性蛋白酶、复合蛋白酶、碱性蛋白酶和风味蛋白酶分别对其进行酶解,以酶解产物的ACE抑制活性和降解效果为指标,筛选出制备海参内脏活性肽的最适工具酶。探究温度、时间、pH、加酶量和料液比等因素对酶解产物ACE抑制活性的影响,通过L9(3~4)正交试验优化酶解工艺。结果显示,最适用酶为碱性蛋白酶,最优工艺为温度55℃,反应时间1h,pH值8.0,加酶量1.5%,料液比1:2,此时酶解液对ACE的抑制率达到87.94%,水解度为23.57%。再经超滤膜将酶解产物分成大于10000u、3000~10000u、小于3000u三个组分,分别测定三部分的ACE抑制活性,结果显示小于3000u组分的活性最高,其IC_(50)为1.2mg/mL。本研究为实现海参加工副产物的高值化利用提供了理论依据。
Sea cucumber visceral which have larger food value is the waste of sea cucumber processing industry.Neutral protease,protamex,alkaline protease and flavourzyme were found to be more suitable for the hydrolysis oiSea cucumber visceral based on ACE inhibition rate and the degradation.ACE inhibition was taken to Alkaline protease has shown efficient hydrolysis for sea cucumber visceral protein to obtain the hydrolysates possessing the highest ACE inhibition rate and its optimum hydrolysis conditions were determined by orthogonal experimental design method as follows;temperature 55℃,time lh,pH value8.0,with enzyme dosage of 1.5%and ratio of matieral to waterwas one to two,in those conditions,the ACE inhibitory activity was 87.94%and the degree of hydrolysis was 23.57%.Peptides withmolecularmass less than 3000,3000- 10000 and more than 10000 u were separated by ultrafiltration,thepeptide with molecular mass less than 3000 u showed the highest inhibitory activity with IC_(50) are 1.2mg/mL.The present study estabhshed a base for effective utilization of sea cucumber processing byproducts.
