虾夷扇贝生殖腺酶解物-核糖美拉德反应产物的抗氧化特性研究

摘要
本文选用碱性蛋白酶制备虾夷扇贝雌性生殖腺酶解物,再与核糖进行美拉德反应获得虾夷扇贝雌性生殖腺酶解物-核糖美拉德反应产物。采用DPPH、ABTS自由基和亚油酸过氧化体系,评价美拉德产物的抗氧化能力,同时对不同反应阶段各指标的变化进行探究,并探讨挥发性成分与抗氧化活性的关系。结果显示,虾夷扇贝雌性生殖腺经碱性蛋白酶酶解3h,其水解度及肽得率分别达24.16%和20.79%。通过比较半抑制浓度(IC_(50))得出,美拉德产物的抗氧化能力较酶解物及核糖本身均显著提高,但弱于抗氧化剂2,6-二叔丁基-4-甲基苯酚(BHT)。相关性分析显示,中间产物、荧光物质及褐色物质与抗氧化活性均呈现正相关,且关联程度高。经GC-MS分析得出,挥发性物质糠醛含量最多,其可能对MRPs的抗氧化活性有一定的影响。以上研究结果说明,经美拉德反应修饰可提高虾夷扇贝生殖腺酶解物的抗氧化能力,具有潜在的开发价值。
In this paper,the hydrolysates fromscallop {Patinopecten yessoensis) female gonad were prepared by usingalcalase.Maillard reaction productswere further obtained from the system of riboseand hydrolysates from scallop(P.yessoensis) female gonad.The antioxidant capacities of Maillard reaction products were evaluated by DPPH 、 ABTS radical scavenging assay and linoleic acid peroxidation assay,meanwhile the index changes of different reaction stage also been explored.In addition,the correlationbetween the antioxidant activity and volatile compounds has also been discussed.The results showed that the hydrolysis degree and TCA soluble peptide yieldof scallop(P.yessoensis) female gonad reached 24.16%and 20.79%,respectively,by hydrolysiswith alcalasefor 3h.By comparing the half inhibition concentration(IC_(50)),the antioxidant capacity of Maillard reaction products was significantly enhanced as comparison with the hydrolysates and ribose,but weaker than butylated hydroxytoluene(BHT).The intermediate product,fluorescent substance and brown substance were positively correlated with the antioxidant activity.In addition,furfural is the most volatile substance in the Maillard reaction productsby GC-MS,which may be related to the antioxidant activity.The results suggest that the antioxidant capacity of scallop female gonad hydrolysates can be improved bv Maillard reaction,having potential value forexploitation.
