萘二甲酰亚胺为骨架的荧光探针在活细胞溶酶体中选择性和特异性检测谷胱甘肽

摘要
谷胱甘肽,经历许多细胞过程,在人体的生理防御系统中起着非常重要的作用~1。因此发展一个有效的方法来监控活细胞中谷胱甘肽的水平具有重大意义。在本文中,我们报道了一个新颖的以萘二甲酰亚胺为骨架,以磺酰胺单元为硫醇反应位点的荧光探针,它能够从半胱氨酸和高半胱氨酸中有效区分谷胱甘肽。生物成像研究表明它在活细胞中具有高度选择性,可对溶酶体中谷胱甘肽水平可视化监测。值得一提的是,不同官能团连接在酰亚胺单元上会影响探针对硫醇的选择性和反应动力学。
Glutathione(GSH),undergoing many cellular functions,plays an important role in the body's biochemical defense system.~1 Developing an efficient method to monitor the level of GSH in living cells is significant.In this work,we report a novel naphthalimide-based fluorescent probe by employing a sulfonamide unit as a thiol-responsive group.It is capable of efficiently distinguishing GSH from cysteine and homocysteine.Bioimaging shows that it has highly selectivity in living cells and can visualize the level of GSH in lysosome.It is worth mentioning that the different group on the imide unit can affect the selectivity and reaction dynamics of probe towards thiols.
