甲酸酯型肽脂质体的氨基酸数量对基因转运性能的影响

基因治疗作为一种革命性的治疗手段,对免疫缺陷病、肿瘤等多种疾病的疗效已经得到了充分验证。阳离子肽类脂作为一种重要的非病毒基因载体,在基因转运方面具有十分广阔的应用前景。近年来研究发现,类脂的结构对基因转运性能具有一定的影响。本研究以课题组合成的不同数量的氨基酸为头部的氨基甲酸酯型肽类脂作为主要载体,考察其粒径、Zeta电位及形态。用激光散射粒度仪检测其粒径和Zeta电位,粒径基本范围在50~200nm,Zeta电位在40~100mV左右。将其与荧光素酶基因(pGFP-N1)结合,形成脂质体/DNA复合物载入宫颈癌细胞(Hela)、肺癌细胞(H460),检测其对细胞的转染效率及毒性。结果表明,氨基酸数量为3个时,荧光素酶基因的转染效率最高,细胞毒性也最低,且与DNA的最佳转染比例范围为2:1~4:1。用MTT法检测毒性表明只有N/P大于4/1时,对细胞的存活率才有一定的影响。本研究将对阳离子类脂的结构设计提供了良好的研究思路及发展方向。
Gene therapy as a revolutionary treatment method for treat a few diseases,such as immune deficiency diseases,cancer and other diseases,has been fully validated.Cationic peptide lipid as an important non-viral gene vectors,has a very broad application prospects in terms of gene transfer.In this study,we varied the number of amino acids in carbamate-linked peptides for gene delivery.They were characterized for their size and zeta-potential by using transmission electron microscope and laser light scattering particle size analyzer.The average particle sizes of these liposomes were from 50 to 200 nm,and their zeta potential was between 40 and 100 mV.Carbamate-linked peptides were used for gene delivery,and obtained the highest transfection efficiency of luciferase gene and the lowest cytotoxicity when peptides had three amino acids.The optimal ratio range of lipid/DNA was 2:1~4:1 and only N/P was higher than 4/1,which had a certain effect on the cell viability by MTT cytotoxicity assay.
