高效液相色谱-二极管阵列法测定人血浆中左乙拉西坦的浓度

摘要
目的:建立HPLC法测定血浆中左乙拉西坦血药浓度的方法。方法:采用甲醇沉淀蛋白,安捷伦Eclipse-XDB-C18(4.6mm×250mm,5μm)柱分离,以0.05M磷酸二氢钾缓冲液(PH=3.2):乙腈=(94:6,v/v)为流动相,二极管阵列检测器检测,紫外205nm波长检测。结果:线性范围为1.0~500μg/mL,3个不同水平添加回收率及相对标准偏差RSD(n=6)为95.5%~97.4%和1.0%~2.3%。日内精密度与日间精密度均小于5%,重现性好。方法的检出限为0.3mg·L~(-1),定量限为1.0mg·L~(-1)。结论:该方法准确,简便,精密度高,可用于左乙拉西坦血药浓度监测。
Objective:A simple HPLC method was established for the determination of Levetiracetam concentration in plasma.Methods:The Levetiracetam was extracted from the plasma samples with solvents of methanol.The extract was separated by Agilent Eclipse XDB-C18 with a mobile phase consisting of tribasic potassium phosphate buffer-methanol(94;6,v/v) and detected by diode array detector.The detection wavelength was at 205 nm.Result:The calibration curve was linear within 1.0 ~500μg/mL,The average recoveries and RSD ranged from 95.5%to 97.4%and 1.0%to 2.3%,at three spiked levels.Inter-day and intra-day RSD were 1.1%~ 1.8%and 1.0%~ 3.3%.respectively.The limit of detection(LOD) of the method was 0.3mg · L~(-1),the limit of quantity(LOQ) was 1.0mg · L~(-1).Conclusion:The proposed method appears to be a rapid,convenient and accurate method to monitor the Levetiracetam concentration in serum and can be used for its pharmacokinetic study.
