人细胞系激发试验(h-CLAT)评价化学物致敏性的研究

摘要
采用体外培养的人急性单核细胞白血病细胞株THP-1进行人细胞系激发试验(h-CLAT),探讨该方法在化妆品原料皮肤致敏评价中的应用。选用8个已知皮肤致敏特性的参照物处理THP-1细胞,根据共刺激分子CD86和CD54的诱导表达情况,判定受试物是否具有潜在皮肤致敏性。结果显示,当以致敏物处理细胞后,THP-1细胞CD86和/或CD54表达明显上调,而非致敏物处理后,CD86和CD54的表达变化不明显。以细胞活力≥50%,RFI CD86≥150和/或RFI CD54≥200来判定是否为致敏物。结果提示,h-CLAT能准确区分致敏物和非致敏物。因此,h-CLAT能够较准确地预测化学物的皮肤致敏性,可用于化妆品原料皮肤致敏性的快速初筛。
Establishing human cell line activation test(h-CLAT) with THP-1 cell line,a human monocyte cell lymphoma,to evaluate the application of this in vitro method on cosmetic ingredient safety assessment.After exposure of 8 reference chemicals to THP-1 cell,skin sensitization potency of these chemicals was evaluated according to the inductive expression of co-stimulatory molecules,CD86 and CD54.A chemical will be considered as an allergen if relative fluorescence intensity(RFI) of CD86≥150 and/or CD54≥200with cell viability more than 50%.With these acceptance criteria,all the chemicals could be classified as allergen or non-allergen correctly.It can be concluded that h-CLAT,a valuable in vitro model for the identification of potential sensitizers,can be used in screening of cosmetic materials.
