miR-202在斑马鱼性腺发育与分化过程中起重要调控功能

miR-202是与细胞分化相关的let-7家族成员,其序列和功能在脊椎动物物种间具有高度保守性。miR-202与哺乳动物性腺的发育相关,miR-202在斑马鱼性腺发育与分化和功能行使上的调控功能尚未见报道。本研究利用CRISPR/Cas9基因编辑系统对斑马鱼胚胎的miR-202基因所在区域进行了片段敲除。结果发现F1代杂合子斑马鱼与野生型对照组斑马鱼胚胎发育过程无明显差异,成熟F1代杂合突变体中雌雄比例并没有明显变化,说明单个miR-202的缺失并不影响斑马鱼性别分化。进一步研究发现,在miR-202敲除的F2代中,野生型,杂合子,和纯合子比例不符合孟德尔遗传定律,其中纯合子比例远小于总数的1/4。成熟F2代杂合突变体中雄性个体比例明显大于雌性个体。本实验初步探究了miR-202在斑马鱼性腺发育与分化及配子发生过程中起到的至关重要的调控作用。该研究为阐述miR-202调控功能,深入了解miR-202对生物进化过程中的作用奠定了基础。
miR-202 is a member of the let-7 family associated with cell differentiation, whose sequence and function 、 gene location and expression patterns are highly conserved across vertebrate species. miR-202 is involved in mammals' gonad development, the function of miR-202 in the gonad development and differentiation of the zebrafish is still unclear. In our research, miR-202 coding region was deleted using CRISPR/Cas9 system. There is no significant difference of embryonic development processes observed between the F1 heterozygotes and the wild type zebrafish zygotes. There is no evident sex ratio change in mature F1 heterozygotes. The genotype of F2 doesn't conform to the Mendel's genetic law, homozygotes are less than 1/4. In mature F2 heterozygotes, the number of male zebrafish is significantly more than the female,indicating that miR-202 is required for the zebrafish gonad development and sexual differentiation.
