gsdf在青鳉鱼性腺发育过程中的功能研究

摘要
gsdf是TGF-β超家族的一个新成员,目前已有研究表明gsdf在青鳉中主要与性别分化有关,但是,在脊椎动物精巢发育过程中的作用机制尚不清楚。为了明确gsdf在青鳉精巢发育过程中的相关功能,本研究通过EGFP-gsdf进行组织分布和个体发生分析,发现在青鳉鱼发育过程中gsdf主要在性腺表达,说明gsdf与性腺的发育密切相关。免疫组化结果表明gsdf在垂体中出现表达,说明gsdf的表达可能受垂体调控,并调控性腺的发育。在青鳉鱼中表达EGFP-gsdf的研究表明,gsdf在胚胎发育早期表达,而在后期发育中消失。后续研究将探讨精巢支持细胞及间质细胞的标记基因(dmrt1;star and cyp17a1)的表达模式,以及雌激素调控基因hsd3b1及睾酮素11-KT与gsdf之间的调控机制。该研究结果表明,gsdf与青鳉鱼性腺发育密切相关,但其调控模式仍有待进一步研究。
gsdf(gonadal soma derived factor) is a new member of TGF-β superfamily. Previous studies had showed that gsdfmainly related to sex differentiation. However, the potential role of gsdf invertebrates of testicular development processremains elusive. In order to identifyfunction gsdfduring testis development in medaka.Tissue distribution and ontogeny analysis revealedthat gsdf mainly expressed in gonads and closely related to the development of gonad.Immunohistochemistry analysis demonstrated that gsdf expressed in pituitary and regulate the development of gonad.Over-expression of EGFP-gsdf resulted in the expression of fluorescence in early phase,but it disappeared in the latter stage.Further research will explore the expression pattern of Sertoli cells and Leydig cellmarker genes(dmrt1; star and cyp17a1),regulation mechanism of estrogen regulated gene HSD3B1 and testosterone 11-KT. The results of the research indicate thatgsdf and medaka gonad development are closely related, but its regulation remains to be further studied.
